Objective: To observe the changes of inducing nitric oxide synthase (iNOS), c-fos protein and bcl-2 protein in myocardial tissue following liver ischemia-reperfusion injury and evaluate the contribution of three factors to cardiac muscle cell apoptosis.Methods: A model of liver ischemia-reperfusion injury was established by imitation. 48 healthy rats were randomly divided into 6 groups as following(n=8): the control group (the group of sham operation), simply ischemia 30 min without reperfusion (I group), reperfusion following ischemia 30 min (I/R group), 1 hr reperfusion following ischemia 30 min (I/R1h group), 2 hrs reperfusion following ischemia 30 min (I/R2h group) and 4 hrs reperfusion following ishcemia 30 min (I/R4h group). The contents of GPT, AKP, γ -GT in the serum were detected respectively. Liver tissue and myocardial tissue were observed by HE staining. The expressions of iNOS and c-fos protein and Bcl-2 in myocardial tissue were examined individually by immunohistochemical technique in each group. The changes of myocardial apoptosis were measured by TUNEL labelling techniqueResults: Liver ischemia-reperfusion induced the remarkable hepatic cell injury. It was found that hepatic cell edema, inflammatory cellular infiltration, even cellular necrosis, Correlated enzyme in serum increased.The data of iNOS was obviously statistic significance in I/R group and I/R4h group(P<0.01). The contents of c-fos protein increased in I group and reached to the peak in I/R4h group. And the expression of Bcl-2 protein in myocardial tissue was statistic significant(p<0.01). The index of apoptosis in cardiac muscle cells showed increasing during different phase of liver ischemia-reperfusion injury (p<0.01). The inflammatory infiltration of the neutrophilic granulocyte, cellular edema and interstitial bleeding were found in myocardial tissue by observing myocardial HE staining sections.Conclusion: The injury of myocardial tissue and cardiac muscle cell apoptosis may be induced following the process of liver ischemia-reperfusion injury. iNOS, c-fos and bcl-2 showed increasing and may be involved into the mechanism of cardiac muscle cell apoptosis. Bcl-2 may play the role of the anti-myocardial apoptosis during the period of liver ischemia. The index of apoptosis in cardiac muscle cells is increasing, but its function of anti-myocardial apoptosis is decreasing .
