Objective: To study the effect and mechanism of transcoronary implantation of bone marrow stromal cells for treating myocardial infarction.Methods: Twelve healthy experimental dogs were randomly devided into 2 groups(6 animals in each group).The bone marrow stromal cells (BMSCs) were procured from the posterior superior iliac spine of the group 1 (transplantation group)animals, cultured, treated with 5-aza, and labeled by BrdU. After the left anterior descending arteries (LAD) were ligated in the both group animals, the cells were autologously infused into the group 1 animals through the distal ligated LAD; the group 2 animals, used as controls, were infused equivalent cell-free culture medium in the same way. The ischemic specimens were harvested for immunohistochemical study 4 weeks after operation. In addition, UCG was used to detect the heart function 24 hours after the operation and before harvesting specimens.Results: The BrdU labled BMSCs could be found at the infarcted zones in all the animals of group 1, and the transplanted cells disseminated all over the infarcted zones. No labled cells could be found at those of control group animals. Echocardiography indicated that the heart function was improved significantly after cell transplantation in group 1, ejection fraction (EF), stroke volume (SV) and cardiac output (CO) increasing 0.085±0.015(p﹤0.01) , 9.167±1.471(p﹤0.01) and 0.75±0.084(p﹤0.01) respectively, compared with those of control group.Conclusions: BMSCs can migrate into and disseminate all over the infarcted myocardial zones after transcoronary implantation, living there, thus ameliorate the heart function.
