Objective : The MCM-2 ( Minichromosome maintenance-2 protein) plays an important role that adjust DNA unwindase in replication of eucaryotic DNA . It only expresses in proliferated cells. It is a novel marker of proliferated cells . Now there is not domestic study about the expression of the MCM-2 in skin.This study detected the expression of the MCM-2 in different regional normal skin,the lesions with benign and malignant hyperplasia by immunohistochemical SP method . To explore whether MCM-2 could be one of the valuable markers for dermatologists to diagnose various kinds of benign and malignant hyperplasia skin disease. MCM-2 was a component of prereplication compound . It playes a very important role in adjusting DNA untwisting enzyme in eucaryotic cell , ensured DNA replication only once in every cell replication cycle, and only expressed in proliferative cells . As a result, MCM-2 could act as a precise marker of proliferative cell . Method: The lesion group I included fifteen patients with Bowen's disease or well-differentiated Squamous cell carcinoma (I class or II class), they were finally diagnosed pathologically. There were six patients with Bowen's disease , and nine patients with well-differentiated Squamous cell carcinoma. The lesion group II included twenty-nine patients with Pseudoepitheliomatous hyperplasia or papillomatous hyperplasia ( finally diagnosed pathologically: chromomycosis 4 cases, sporotrichosis 2 cases, seborrheic keratosis 5 cases,verruca vulgaris 4 cases, chronic eczema 4 cases, dermatofibroma 4 cases, non-specificity dermatitis 6 cases). The normal control group included 37 healthy adult's skin. The samples were taken from face ( 7 cases), limbs( 9 cases ),armpit( 8 cases ),trunk( 8 cases),metacarpus or metatarsus ( 5 cases ). All lesion samples were paraffin block file from department of dermatology,second hospital of Hebei medical college, from January 1998 to June 2004 , and they were all made definite pathological diagnosis. All normal samples were superfluous skin obtained by operation and were followed the standard processing to make paraffin sections. The distribution and intensity of expression of MCM-2 in the epidermis of the samples from well-differentiated Bowen's disease or Squamous cell carcinoma(lesion group I), Pseudoepitheliomatous hyperplasia or papillomatous hyperplasia(lesion group II) and health adult (normal control group) were detected by immunohistochemical SP method . Results1.The distribution of sex and age: In the lesion group I , male 10 patients(66.67%) and female 5 patients(33.33%) , the oldest patient was 80 years and the youngest was 40 years, average age was 66.76±15.36 years. In the lesion group II , male 17 patients(58.62%) and female 12 patients(41.38%) , the oldest patient was 78 years and the youngest was 30 years, average age was 66.25±14.37 years. In the normal control group, male 23 patients(62.16%) and female 14 patients(37.84%), the oldest patient was 70 years and the youngest was 55 years, average age was 65.52±16.65 years. 2.The part of MCM-2 expression: The distribution of MCM-2 positive cells in lesion group I ,lesion group II and normal control group was different. In the lesion group I , MCM-2 expressed not only in the cells of stratum basalum but also in the cells of stratum spinosum and above it. In the lesion group II MCM-2 expressed in the cells of stratum basalum and in the a small quantity cells of stratum spinosum. In the control group, MCM-2 expressed only in stratum basalum'cells. 3.The intensity of the MCM-2 expression in the epidermis: The intensity of the MCM-2 expressed in the epidermis of the samples from the lesion group I ,lesion group II and normal control group was different. In the lesion group I : significantly positive lesion 8, positive lesion 5, weakly positive lesion 1, negtive lesion 1; In the lesion group II : significantly positive lesion 3 ,positive lesion 18, weakly positive lesion 6,negtive lesion 2; In the normal control group: positive lesion 1, weaklypositive lesion 30, negtive lesion 6.
