OBJECTIVE To find out the best method of observation of culture cells under AFM. Normal cells and tumor cells were observed by AFM in order to find out the difference between their membrane structures. We also exposed cells to different temperature for different period of time and obtained their AFM pictures and we explore the heat-induced cells membrane structure changes and their meaningsMATERIALS AND METHODS We chose Fb cell, Hela cell, MCF7 cell, Ma-Fb cell, DMEM (Sigma, Inc. USA) +10%(v/v)deactivated Similar Standard FBS (Lanzhou National HyClone Bio-Engineering Co., LTD), 0.25% Trypsin(1:250, Amresco Inc.), 2% formaldehyde solution, citrate buffer and 5% dextrose solution.To find the AFM specimen preparation: cells be fixed by 2% formaldehyde solution and dried at room temperature; cultured cells were cleaned by 5% dextrose solution or citrate buffer and dried at room temperature.The preparation of culture cells specimen: inoculated cells in cell culture flasks with cover glass in, regularly cultured the cells until they covered 80% of the flasks' bottom area, took out the cover glass and fixated it by 2% formalin solution, then dried it at room temperature; The sample preparation of heated culture cells: exposed the culture cells at different temperature for different period of time, then took out them, made them into suspension, washed the cells with 2% formalin 1000rps,3 times.
