Objectives:1. To explore the mechanism of sex differences in EAE.2. To explore the effect of Extacts of Medicinal herbs for EAE. Methods:1. Male and female SJL/J mice were injected s.c. with PLP139-151 and CFA. Clinical symptoms were assessed daily.Draining lymph nodes were harvested separately from male and female mice at 10 days post-immunization. The lymphoid cells were cultured separately in tissue culture plates in stimulation medium with PLP. Then, T cell proliferation to PLP was observed by MTT ; IFN—γ and IL-4 were measured by ELISA.2. Female SJL/J mice were immunized with PLP139-151. At 10 days post-immunization, lymphoid cells were cultured in a flat-bottom tissue culture plate in stimulation medium containing PLP with or without Extacts of Medicinal herbs at varying concentrations. Then, T cell proliferation to PLP was observed by MTT; IFN- γ and IL-10 were measured by ELISA.3. Female SJL/J mice were immunized to induce EAE. After 6 days post-immunization, the first group was given Extacts of Medicinal herbs every other day through celiac injection; the second group was given prednisone every day through celiac injection; the third group served as control group. Then, all mice were assessed daily for clinical signs of EAE. Results:1. The onset time of female SJL mice was 15 ± 2.1 days less than that of male ones (22 ± 4.3d) .
