To investigate the dose -effect and time - effect relationship of Sodium selenite and All - trans Retinoic acid on changes of optical density , cell cycle and mitosis index in lung cancer cell line A549.MethodsThe lung cancer cells A549 were cultured and treated by Sodium selenite, All - trans Retinoic acid, Sodium selenite combined All - trans Retinoic acid at the concentration of 2. 5 (xmol/L, 5. 0-μmol/L, 10.0μmol/L and 20.0μmol/L, respectively, and at different time interval . The MTT colorimetric assay was used to analyze the optical density. Flow cytometry (FCM) was used to detect the percentage of cells at G0/G1phase, S phase and G2 + M phase . Taked cells fixated and dyed , counted mitotic cells with microscope, then worked out mitosis index. A total of five groups were designed, the blank control, 0.095% volume ethyl alcohol control , Sodium selenite groups, All - trans Retinoic acid groups, Sodium selenite combined All - trans Retinoic acid groups.ResultsThe optical density of Sodium selenite, All - trans Retinoic acid groups of 2.5μmo/L - 20. 0μmol/L and combination of the two drugs at the same doses as above groups that had treated over 24 hours - 96hours was significantly high-...
