Objectives: We established an animal experimental model of hyperthyroidism related muscle disease by giving L-thyroxine sodium salt(T3) by intraperitonealing to the healthy female Wistar rats. Then give xiaoyingqiangjitang to the rats by injecting to their stomach. The morphological changes of skeletal muscle cells and motor end plate were observed by light and electron microscopy. The changes of enzyme activity of Succinate dehydrogenase (SDH) in skeletal muscle cell and Acetylcholin esterase (AchE) in motor end plate were showed by enzyme cytochemisty, then observed by light and electron microscopy. And the mechanisms of treatment caused by xiaoyingqiangjitang were investigated.Methods: The thirty-five healthy female mice were randomly divided into seven groups: the control group ;given intraperitoneal injection for 28 days and for 42 days; given intraperitoneal injection for 28 days and for 42 days then treated with xiaoyingqiangjitang and normal solute for 30 days respectively. The control group was given saline solution injection for the same duration as T3-treated group. The T3-treated group were given intraperitoneal injection L-thyroxine sodium salt, 250μg/lml, 1ml/100g once a day. Then the skeletal muscle were removed and processed by following procedures. Some specimens were made into paraffin section, stained by hematoxylin-eosin(HE) staining method and morphological changes were observed by light microscope. Some specimens were prepared into transmission electron microscope (TEM) sample and the effect of xiaoyingqiangjitang on skeletal muscle ultrastructural alteration was observed by TEM. For enzyme cytochemistry, other specimens were sliced into 15μm and 40μm frozen sections by cryostat microtome. The 15μm sections were stained by SDH and AchE, and the enzyme activity was observed by light microscope and image analysis technology was employed to study the quantity changes. The 40μm sections were incubated in AchE incubation solution, then prepared to TEM samples and the enzyme activity changes of AchE on motor end plate were observed by TEM.Results : (1) Morphological observation shows: There were no remarkable abnormities of skeletal muscle cell observed by HE staining except for more nuclears under sarcolema after injecting T3 for 28 days; but for 42 days there were more abnormalities: the space between fibers become slight widen, the cross striatum become faint. Corpulent muscle fibers and atrophic ones were alternate. After be givien Chinese medcine, all abnormities disappeared basically. And under TEM, we found that the myofilaments in T3-treated groups arranged irregularly, for 28 days, some vacuole could be observed. Some sarcomere shorten. Some mitochondria swelled, some lengthened. For 42days, the cristae of mitochondria ruptured, inosculated and even disappeared and some mitochondria became bulgy and nonuniformed size.
