Objective Grape procyanidins(GPC) are natural polyphenols extracted from grapes.It was reported that GPC have a lot of bioactivities such as antioxidation,antiradical and antitumor effects.In this study, the effects of grape procyanidins on the proliferation of mice pancreas cells were observed in vitro and vivo.Methods 1.In vitro experiment, The pancreas cells of mice were pretreated and not pretreated by GPC and then were cultured with different concentration of glucose and H2O2 respectively for 24 hours.The proliferation of normal and injured pancreas cell was determined by MTT assay.2. In vivo experiment, Different dose of GPC was given to the mice by mouth who radiated and not radiated by 60Co- γ -ray, then proliferation of pancreas cell was measured by MTT, Flow cytometry was used to analysis apoptosis of the mice pancreas cells, microstructure of pancreas cells were observed by microscope. Immunohistochemistry method was applied to measured the expression of bcl-2 and bax protein in mice pancreas cells.Results 1.Vitro experiment.The cell proliferation of H2O,30mmol/L glucose and 2.0mmol/L H2O2 groups not pretreated by GPC were 0.102±0.008, 0.060±0.002 and 0.053 ±0.006 respectively, lower than that in groups pretreated by 100 μ g/ml GPC. The differences above were significant in statistics( p<0.05)2.Vivo experiment:2.1 Acute radiation experiment,The cell proliferation rate and bcl-2 expression of high dose GPC protective group were 3.16%± 0.13% and 49.8% respectively, higher than that of radiation control group,but the cell apoptosis rate and bax expression were 36.65% and 55.0% respectively, lower than that of radiation control group.All the difference above were significant in statistics( p<0.05).2.2 Chronic radiation experiment, The bcl-2 expression and cell proliferation rate of GPC protective group were 51.1% and 4.89%±0.16% respectively, higher than that of radiation control group,but bax expression was lower than that of radiation control group.Cell microstructure injury was lighter than than of it.All the difference above were significant in statistics (p<0.05).Conclusions 1 .GPC can promote the proliferation of normal pancreas cells.2.GPC has protection on the proliferation of pancreas cells injured by radiation,high glucose and H2O2.3.This study will offer scientifical theory to treat and prevent some pancreas diseases.
