Objective To observe the functional states of Th1/Th2 in peripheral blood mononuclear cells(PBMC) of asthmatic children ,and to study the effects of Utilin'S on functional states of Th1/Th2 in PBMC of asthmatic children .Methods 20 asthmatic children and 15 healthy children controls were accessed.After stimulation to PBMC of asthmatic children and PBMC of healthy children with PHA ,PHA+Ul(1×10-2mg/ml),PHA+U2(1×10-3 mg/ml),respectively,and cultured for 48 hours in vitro,the levels of IFN-r and IL-4 in the supernatants of PBMC were detected by ELISA respectively.Results (1)After stimulation with PHA , the level of EFN-r produced by PBMC of asthmatic children were statistically lower than that of the healthy children's PHA group(t =3.05,p<0.01),but the level of IL-4 produced by PBMC of asthmatic children were statistically higher than the healthy children's PHA controls(t'= 11.18,p<0.01),and the ratio of IFN-r/IL-4 of asthmatic children was lower than that of the healthy children's PHA controls(t= 4.16,p<0.01). (2) After stimulation with PHA+U1, the levels of IFN-r and IL-4 in PBMC of asthmatic children were significantly higher than those of the group of healthy children (t' =14.06,10.92, p<0.01, respectively). The ratio of IFN-r/IL-4 of Ul group of asthmatic children increased than that of the Ul group of healthy children (t' =3.79,P< 0.01); The levels of IFN-r , IL-4 and the ratio of IFN-r/IL-4 of Ul group of asthmatic children increased than those of the PHA group (q=31.76,7.34,12.34, P 均 < 0.01); after stimulation with PHA+U1, the level of IFN-r in PBMC of healthy children were markedly higher than that of the PHA group of healthy children (q=4.12,P < 0.05),but there was no difference in I1-4 level and the ratio of IFN-r/IL-4 between two groups(q =1.57,1.28,P > 0.05 respectively).(3) After stimulation with PHA+U2, the levels of IL-4 in PBMC of asthmatic children were significantly higher man those of the group of healthy children (t'=3.94,p<0.01).Though the level of IFN-r and the ratio of IFN-r/IL-4 of U2 group of asthmatic children increased than that of the U2 group of healthy children , there was no difference between two groups(t= 0.38,0.15,p>0.05 respectively); After stimulation with PHA+U2, the levels of IFN-r and the ratio of IFN-r/IL-4 in PBMC of asthmatic children were significantly higher than those of the PHA group , (q= 10.35,7.38,P < 0.01, respectively), But the levels of IL-4 lower than that of the PHA group (q=3.29 P < 0.05); And the levels of IFN-r, IL-4 and die ratio of IFN-r/IL-4 of U2 group of asthmatic children lower than those of the U1 group (q=21.41,10.62,4.%, P < 0.01 respectively); with no difference between healthy children U2 group and the PHA group(q=0.74,0.35,0.35, P 均 > 0.05 .respectively), only the levels of IFN-r, the difference could be found between healthy children U2 group and Ul group (q=3.38, P < 0.05), but IL-4 and IFN-r /IL-4 were no difference could be found between healthy children U2 group and Ul group (q=1.21,0.93, P>0.05 , respectively).Conclusion (1)The study shows that the Th2 cells hyperfunction,and the Th1 cells inactiv-...
