Objective: To detect the overexpression of serum P53 protein and heat shockprotein 70(HSP70) in peripheral blood lymphocytes of workers exposed to cokeoven emissions and to study the correlation between P53 and HSP70 ,Byanalyzing the increased risk in P53 gene mutution and the overexpression ofHSP70,So as to provide more powerful theory gist for early health surveillanceof coke oven workers.Methods: 118 coke oven workers were chosed as the exposing group and 50health people without any occupational exposure history of carcingen werechosed as the control group.Some information was investigated includinggeneral information,history of occupational exposure and respiratory disesesetc. by uniform questionnaires which were inquiried by some people that weretrained. The peripheral blood --2ml was drawn to separate serum and 3ml wasused for seperating lymphocytes,Western blot was used for determing the levelof P53 and HSP70.Serum P53 and HSP70 and respiratory diseses werecompared between two groups.Some affecting factors of serum P53 and HSP70and the correlation of serum P53 and HSP70 were analysed.Results: 1. 15 cases with positive serum P53 and 18 cases with positive HSP70in peripheral blood lymphocytes were found mostly in more than 20 yearsexposed group in 118 cases coke oven workers.The positive detection rates ofP53 in the exposed group(12.82%) were higher than the control group(2.00%,P>0.05); the positive detection rates of HSP70 in the exposed group(15.38%)were higher than the control group(2.00%,P<0.05).But the positivecoexpression rates of serum P53 and HSP70 was 11.02%. 2. Compared with the control group, P53 and HSP70 levels in thedifferent exposued group increased significantly(P<0.05),and the expression ofP53 and HSP70 increased gradually in the exposued group with years of service(r_s 0.920,0.931 respectively,P<0.01). 3. Compared with the control group , P53 and HSP70 levels in thedifferent work sites increased significantly(P<0.05),however the significancebetween in the different work sites was no evidently difference. 4. In the exposed group ,the levels of HSP70 in peripheral bloodlymphocytes had strong positive correlation with serum P53 (r=0.874,P<0.01). 5. The incidences of chronic rhinits and pharyngitis were 32.2%,37.3%and 16.0%,12.0% respectively in the exposed group and the control group. Thedifferentia between two groups was significant. 6. The mostly risk factor of increasing serum P53 and HSP70 inperipheral blood lymphocytes was the exposed time.Conclusion: The expression of serum P53 and HSP70 in peripheral bloodlymphocytes and the incidences of chronic rhinits and pharyngitis in coke ovenworkers were higher than general people. Serum P53 and HSP70 wereincreased with the time prolonged. The exposed time were mostly risk factor ofincreasing serum P53 and HSP70.The coexpression of HSP70 and P53 in thecoke oven worker and the strong positive correlation between HSP70 withserum P53 suggested that HSP70 and P53 may have associated expressiontendency. In conclusion, the results suggested that COE was likely to causemutation of P53 gene and induce to the overexpression of heat shock proteins(mostly HSP70) and that HSP70 can regulate the function of antioncogene P53,which increasing the incidence of lung cancer and others diseases.Sodetectining HSP70 and P53 is in favor of early health surveillance of coke ovenworkers.
