Objective: This study was designed to investigate the expression of Cx43 and VEGF protein in Hep-2 affected by Retinoic Acid and inhibition effect of Retinoic Acid on Hep-2. Methods: Inhibition effect of Retinoic Acid on Hep-2 was examined by methyl thiazolyl telrazolium(MTT) assay. Cx43 and VEGF protein in Hep-2 were examined by immunocytochemistry. Results:Growth of cancerous cell is regulated by Retinoic Acid in dose and time manner .After treated with the difference concentration Retinoic Acid,the growth of cancerous cells was inhibited .Inhibition rate of growth on Hep-2 was significant increased with 0.5micromol/l Retinoic Acid and is the peak with concentration of 2micromol/l Retinoic Acid, following a little decreased. There was a significant up-regulation on expression of Cx43 in Hep-2 treated with the Retinoic Acid and down-regulation on expression of VEGF. There are significant difference among each group treated by Retinoic Acid with the one untreated by it about the expresses of Cx43 and VEGF proteins (all of P<0.01). After cancerous cells is treated by RS11237(agnoist to Retinoid X recrptor--RXR) or BMS439(antagonist to Retinoic acid receptor--RAR) for 4 hour ,as follow is treated by Retinoic Acid for two days. There is a significant difference on inhibition rate of growth and expression of VEGF.and Cx43 compared with the control group(P<0.01). Conclusion: Growth on Hep-2 cells can be inhibited by Retinoic acid via Retinoid X recrptor(RXR) or Retinoic acid receptor(RAR) Inhibiting might be relative with up-regulation on expression of Cx43 and down-regulation on expression of VEGF.via activation of Retinoid X recrptor(RXR) or Retinoic acid receptor(RAR)...
