Object:to investigate the activity of MAPK, the production of fibronectin and expression of c-fos in mesangial cells cultured in high glucose and the effect of high glucose on the intracellular Ca2+ concentration of human mesangial cells. To explore the relationships among these phenomena and to explain the mechanism of diabetes nephropathy. To investigate the activity of MAPK and the production of c-fos in human smooth muscel cells cultured in high glucose.to explore the mechanism of diabetic vascular complication. Method:The human mesangial cells and the human smooth muscle cells cultured were stimulated by high glucose. PD98059 was a special inhibitor of MAPK.The MAPK activity was measured by liquid scintillation counting. the expression of c-fos was measured by immunochemistry.The fibronectin production of human mesangial cells was analyzed by ELISA. The intracellular Ca2+ concentration of human mesangial cells was mensurated by fluorescent measure.The proliferation of human mesangial cells were assayed by MTT. Result:Compared with the contorol glucose, the high glucose increased the activity of MAPK and the expressoion of c-fos .When PD98059 used, These two were all decreased in both cells.The production of FN and the intracellular Ca2+ concentration of human mesangial cells were all increased cultured in high glucose. When PD98059 used,the production of FN was decreased.High glucose inhibited the proliferation of human mesagial cells. Conclusion:In human mesagial cells, the activity of MAPK ,the expression of c-fos and the production of FN were increased when treated with high glucose. These three penomenons were correlated with each other.In human smooth muscle cells,the activity of MAPK and the expression of c-fos were increased cultured in high glucose. The production of FN was increased in human mesagial cells when treated with high glucose.The activity of MAPK may affect the production of FN.
