The Effect Of Garlicin On Sodium Channel Current In Isolated Ventricular Myocytes

[Objective] To observe the effect of Garlicin on sodium channel in isolated ventricular cell of rats, to find out the mechanism of antiarrhythmia of Garlicin on the ionic channel level.[Methods] Single ventricular cell of guinea pig was obtained by enzymatic dissociation. The whole-cell patch clamp recording technique was used to record the change of currents of INa by the effect of Garlicin.[Results] (1). Garlicin could decrease the sodium channel current in a dose-dependent and voltage-dependent manner. (2) Garlicin concentration has the impact between current density and voltage of INa, its current density in menbrance voltage was -13.41 ±6.95 pA/pF(n=6) at peak potential of-40mv prior the use of Garlicin. Garlicin 500 μ mol.L-1 decreased current density by 48%(P<0.01) from peak current density was prior -13.41±6.95 vs post -8.53± 3.70 pA/pF, significant value established(n=6, P<0.01). (3) The activity curve had no significant change by use of Garlicin 500? mol/L, their V50 were -44.72±18.61 vs -45.66± -17.86 mV and Slope were -2.7±0.8 vs -2.2±0.7,( n=6, P>0.05). The result of inactive curve revealed the Garlicin can move a shift to the negative direction of its potential statistically, their V50 were -88.61±9.6 vs -103.03± -8.9 mV and Slope were -7.85±0.88 vs -7.55±2.75, (n=6, P>0.05).[Conclusion] Garlicin exercutes the blocked function to INa as dose reliance and voltage reliance, but does not alter the origiNal Ⅰ-Ⅴ curves and stimulating curves.Others garlicin effect the inactive curve of INa obviously and produces the delay in recoveried procedure. Garlicin could block INa, in its iNactivity state by a dose-dependent and voltage-dependent manner.During the inactivity of Ina recovery, the Garlicin made action to inactive cahnnel not to nonresting or active channel, that concluded the Garlicin has the pharmacologic effect to rapid tchyarrhythmia obviously.
