Objective:For a long period of time,we have repairing bone defects created from wound,infection,or other reasons by the method of autograft of bone.However the bone autografting gives priority to osteogenic activity as crawling and substituteing,the process is slow and it is limited by cut place,area and amounts. The use of calcium alginate gelatin which is a perfect tissue engineering bracket material in repairing of orthopaedics wound is a hotspot at the research of tissue engineering.The calcium alginate gelatin looks as 3D reticulation by electron microscope,and is compare to "egg box".At present,it is used widely at pharmacy, foodstuff, bioengineering material.The use in orthopaedics refor mainly to repair of cartilage cell.As cell carrier,it has some characteristics such as:good dynam capability,chemistry stability, low toxicity,low pyrogen,low immunoreaction,sure spacing rate and interval,good consistent quality with cell and the operation could being done in one step,allowing exchange oxygen nutriment and waste,so that it set up good 3D spacing structure for cell growth,have good bone transmit effect.Many scholar prove that the periosteum can ossify by animal experiment and electron microscope.At early of 20th century Axhausen proved the free periosteum can ossify, Many scholar also prove that the ossifying ability of periosteum comes from it's germinative layer at 1970's and they advanced the conception of "acuting desquamation"so that the germinative layer can be protected fully.there is osteoblast desmocyte and osteoprogenitor cell etal.it can create cartelage and bone,the ossifying ability be strong,is better proffer of germ cell and it has self-ossifying ability.the grand old man repaired bone defect with periosteum chip and got good effect,so in this experiment the periosteum is made to cell suspension so that the periosteum can be taked full adventage and there is multi-centre ossifyication, to sum up,the repairing speed is picked up.There is abundant Transforming Growth Factor(TGF-β) and Bone Morphogenetic Protein(BMP). They can stimulate periosteum cell differentiation and periosteum ossifyication (bone induction).We compound periosteum cell suspension in calcium alginate gelatin,bring the osteoacusis the self-ossifyication and the bone induction into play sufficiently.In this experiment,we sum up the research experience of the grand old man,and take stock of whether periosteum cell suspension compound with calcium alginate gelatin can ossify or not in self-entironment and the mode of ossifyication and find a new method which is used conveniently in clinic to therapy bone defect there by. Method:60 adult New Zealand rabbits are divided into 3 group of 20 animals at random.All the animals are operated intostandard bone defect of 15 mm in left radius.Group A are filled with calcium alginate gelatin/ periosteum cell suspension;Group B are filled with calcium alginate gelatin;Group C are filled with periosteum cell suspension.The animals are sacrificed at 2nd,4th,6th,8th and 12th week postoperation,4 animals of each group are sacrificed each time.After muscle are removed away from the specimens,results are revealed in gross observation,histology and X-radiography.Moreover,level of alkaline phosphatase(ALP),serum calcium are measured on each group animals.The complexion of bone forming are observed in macroscopical and microcosmic side,and by the score of X-rays carried through in according to the condition of bone shape,bone regeneration,bone defect's connection and the statistics treatments of ALP and serum calcium,the effect of bone defect repaired with autograft of periosteum cell suspension compound by calcium alginate gelatin has been evaluated objectively. Result:Gross observation,histological analysis,X-ray film and biochemical examination are taken at 2nd,4th,6th,8thand 12thweek postoperation.the result are following:In group A,there are distinctive periosteum reaction and lots of callus growth in transplanting area,consistent callus finally bridging the gaps of defect,new bone contoured...
