OBJECTIVE: To investigate the molecular mechanism of perfluorocarbon (PFC) relieving Inflammatory reaction and the effect on the ability of pulmonary host to defense against microorganiams during acute lung injury, the protein expression, ratio of apoptosis and phagocytosis of alveolar macrophage(AM) were studied by interfering with PFC.METHODS: AM were obtained after sacrifice from Waster healthy male-rats by total lung lavage and purfied by adherent method. AM suspension were divided into 4 groups for intervention: control, LPS, PFC and PFC+LPS. LPS were added with 10ug/ml and PFC with 30% volume to culture solution. After the preincubation for 20h, AMs were resuspensed and intervented for 4h. Then, the protein of 4 groups were abstracted to study the interclass-difference by SELDI-TOF-MS; the AMs were collected and incubated with 1% chicken erythrocyte suspension. slicemaking, Wright's staining were implemented for calculating the ratio and index of phagocytosis, and the disparity of 4 groups were compared with one-factor analysis of variance. After purification, AM were incubated with serum-free medium and intervented for 24h, then collected and observated by transmissional electron microscope and combining flow cytometry to compare the disparity of apoptosis-ratio between groups.RESULTS: 1.
