Objective: To establish hypospadias rat model and study the effectmechanism in gene with Di(n-butyl) Phthalate.Methods: Sixty Pregnant Sprague-Dawley rats were divided randomly into four groups: fifteen conceived rats were respectively divided into hypospadias model group (A1) and experimental detectiongroup (A2 ) with DBP and fifteen respectively with soybean oil forcontrol group B1 and B2. From 12th to 20th day after the day of conception of the female rats , the rats were gavaged with DBP 500mg/(kg.d) inDBP group and soybean oil in the control group and each rat of the garage capacity was 1 ml/day. After all pregnant ratshad delivered, the male offspring were counted and the anal genitaldistance (AGD) and body weight were measured with anatomy microscope. Hypospadias were also observed after postnatal seventy days of themale offspring in A1 and B1 groups. Testicular RNA isolated from three randomly selected GD 20 fetuses per litter in A2 and B2 was used for real-time RT-PCR for the following genes: P450 side-chaincleavage enzyme (P450scc), 3 β -hydroxysteroid dehydrogenase (3 β -HSD),...
