ObjectivesTo study the expression and the role of the PKR in MM. To set up a basement for the treatment that may be more effective by modulating the PKR signaling system in MM.Methods1. The expression of PKR was examined in 42 cases of MM and 25 cases of ordinary nevi by immunohistochemistry. The relationship between the expression of PKR and the clinical features of MM was analyzed by SPSS.2. The expression of PKR, p-PKR and p58 in MM cells and MC cells in vitro was examined by western-blotting.3. The relationship between the expression of PKR and PCNA was analyzed by SPSS.4. The expression of PKR and p-PKR in MM cells in vitro was modulated by IFNα 2b, Poly I:C and 2-AP. The expression of PKR and p-PKR in MM cells was examined by western-blotting and immunofluorescence. The level of apoptosis and proliferation was analyzed by MTT and FCM.Results1. The positive expression rate of PKR in MM is higher than that in ordinary nevi (X~2=55.548, P=0.000 ) .There isn' t significant difference between between the expression of PKR and the clinical features of MM.2. The expression of PKR and p58 in MM cells was higher than that of MC cells in vitro. However, the expression of p-PKR in MM cells and MC cells are both low.3. There is a reverse-correlated expression between PKR and PCNA in MM in vivo (r =-0.442; P=0. 003) .4. In vitro , by the treatment of IFNα 2b, the expression of PKR in MM cells increases significantly, the level of apoptosis increases slightly and the level of proliferation decreases slightly. By the treatment of IFNα 2b and Poly I:C. Poly I:C and 2-AP, the expression of PKR and p-PKR in MM cells increases significantly, the level of apoptosis increases significantly and the level of proliferation decreases significantly. By the treatment of 2-AP, the increase of the expression of p-PKR, the increase of the level of apoptosis and the decrease of the level of proliferation are all inhibited.
