Objective Histone deacetylase inhibitors (HDACIs) can inhibit the proliferation of many kinds of tumor cells and induce the apoptosis and differentiation of them. They have exclusive cytotoxic effect on tumor cells and can cooperate with many chemotherapeutic agents without serious cytotoxic effect on normal hemopoietic cells. Recently, the clinical trials about HDACIs are on the I or II phase. VPA is a traditional medicine used to treat falling sickness. It can be accepted by most patients. Recently, It was found that VPA has the special function of HDACIs in the level of pharmacology and can induce the apoptosis and differentiation of tumor cells. So far, there're no reports about the effects of VPA on acute myeloid leukaemia cells in China and no reports about the effects of VPA on the expression of Racl which plays important role during the cell cycle, apoptosis and differentiation all over the world. In this experiment, we took HL-60 cells as targets and examined the effect of VPA on the proliferation, differentiation and apoptosis of leukemia cells. At the same time, we analyzed the effect of VPA on the expression of Racl mRNA.Methods Growth curves of HL-60 cells were performed by MTT assay.
