Objective:Tongmai oral liquid possesses a good curative effect for chronic nephritis in clinic, which had been proved by Guangdong Province Hospital of TCM for a long time. This paper provides scientific bases for compositive mechanism, clinic application and treatment mechanism of Tongmai oral liquid through such research work respectively: The quality control of materials for Tongmai oral liquid, extracting and purification technology for main chemical parts, screen out pharmacological effect of different chemical parts, effective chemistry part's fingerprint research, separation and identification of main components of Tongmai oral liquid.Methods:On the basis of medical literature, the quality control method for medicinal materials of Tongmai oral liquid was established by using Tin-layer chromatography method to differentiate and spectrophotometry to mensurate the content of polysaccharides and total saponins; single factor investigative method and orthogonal experimental design were used to screen out the extracting process of Tongmai oral liquid. The macroporous resin D101 was used to gather and purify total saponins, the pharmacodynamic effect of different chemical parts of Tongmai oral liquid was investigated with blood rheology index and immunization parameters; the fingerprint of total saponins which had been proved effective was studied preliminarily by using Tin-layer chromatography method and HPLC method; The main chemical components ofTongmai oral liquid were separated and purified by using Column chromatographic technique, Crystallization method etc, Combined with IR, MS,1HNMR, 13CNMR, HPLC method to identify these components.Results:1 The results of quality control method study for medicinal materials in Tongmai oral liquid:In Tin-layer chromatography of Astragalus menbranaceus (Fisch.)Bge. , Astragaloside is yellow blot, Rf value is 0. 35, the Tin-layer chromatography action does well. In the Tin-layer chromatography of Panax notoginseng (Burk.) F. H. Chen, the blots of Ginesenoside Rg, and Ginsenoside Rbt are amaranthine blots, Rf value is 0.15 and 0.37 respectively, the Tin-layer chromatography action does well too. Measuring several batch samples, we found the contents of Astragalus menbranaceus (Fisch. )Bge. polysaccharides are between 5. 56%7.18% ; the contents of Panax notoginseng (Burk.) F. H. Chen saponins are between 6. 96%8. 10%.2 The results of extracting and purifying process study:The condition of extracting process for Tongmai oral liquid was optimize by single factor investigative method and orthogonal experimental design, using the yield and content of polysaccharides as parameters. The optimum extractive process is: soaked for 12 h, extracted twice with 8 times of water,3 h per time. The yield and content of saponins were 2. 18%2. 54 and 68. 5177. 23% respectively by the method of macroporous resin D101 gathering and purifying.3 The results of pharmacodynamic study on different chemical parts of Tongmai oral liquid:The results on immunity function of mice showed that the immunity function of group TM1, group TM2, group ZG and group DT were improved in different degree, (P<0.05-0. 01), while the group TM3's mice hardly affected on the immunity function( P>0. 05).The results on the blood viscosity of rat showed that group TM1, group TM2 and group ZG can decrease the whole blood viscosity and plasma viscosity in different degree (P<0.05-0. 01), while group DT and group TM3's rats haveno changes on viscosity

The group TM1, group TM2 and group ZG can decrease the erythrocyte aggregation index, erythrocyte rigidity index, erythrocyte transmutation index and electrophoresis index (P<0.05-0.01), while group DT and group TM3's rats have no changes on such index (P>0. 05).4 The fingerprint results of effective chemistry parts of Tongmai oral liquid:The results of TLC research showed that there are 10 blots in different groups of Tongmai oral liquid saponins, their Rf value are 0.11, 0.17, 0.19, 0.23, 0.30, 0.33, 0.47, 0.50, 0.58, 0.63 respectively, 0.33 is ginesenoside Rg, and 0. 17 is ginsenoside Rba thereinto, no blot of astragaloside is found in Tongmai oral liquid saponins.The results of HPLC showed that there are 12 common peaks in different groups of Tongmai oral liquid saponins in all, the average relative time are 10.075, 21. 269, 24. 163, 29. 399, 31.283, 38.291, 40.149, 41.148, 42. 927, 52. 400, 53.538, 62. 632. The relative time of 24. 163 is ginesenoside Rg, and 38.291 is ginsenoside Rb,, the sum peak area of them is more than 30% of total area.5 The results of main chemistry components of Tongmai oral liquid:Three monomer compounds (TM-1, TM-77, TM-92)were separated from the Tongmai oral liquid, they had been identified as cane sugar, ginesenoside Rg, and ginsenoside Rb,.Conclusion:The control quality methods of single medicinal materials are convenient and have good repetition, which can be used as the main control quality methods in manufacture and research. The effective parts of Tongmai oral liquid can be extracted more completely in the process that has been selected by orthogonal experiment. This technology suits for manufacture practice. The content of total saponins enriched and purified by macroporous resin D101 is higher that can be investigated as new kinds of materials of traditional Chinese medicine farther. The pharmacodynamic experiments showed that the main efficacious part of Tongmai oral liquid used to treat chronic nephritis is the total saponins and total polysaccharides, the total saponins is more effective than the total polysaccharides, and combined them can create a better...
