Experiment Studies On The Establishment Of Neuroblastoma Vitro Cell Line And The Bear-tumor Mice Model

Objective: Neuroblastoma is one of the most common tumor of children,some of them can grow mature and disappear spontaneously.we use enzyme digestion and erasion to purify neuroblastoma cell line, to establish cell suspens. Further more, we can study the relationship between the biological characters of NB and stage of tumor, diagnosis of patient and spontaneous regression.Method: Use the fresh tumor tissue to establish the in vitro cell line and use the cell line established by ourselves to get the bear-tumor nude mice model.Take the fresh tumor tissue from the patient under the sterilizing situation during the operation, used the enzyme digestion method to establish the in vitro cell line. Used the enzyme digestion method and erasion method to get the single cell suspension.Result: We got the in vitro cell line of primary tumor successfully. Now we got the 52th generation cell line.The way of digestion culture is better than that of tissue culture. We can see the cells digested by enzyme the same time if we culture cells by the way of digestion culture, all of them have the morphology of round shape, they became little shuttle shape the second day. The NB cells have the same shapes approximately, small shuttle shapes; tear shapes; and triangle shapes. All these are infantile cells. Some NB cells that have a long prominence are mature cells. The cell line we founded was identified as NB cell line by a series test.We get the single cell suspension successfully.Conclusion: We have the ability to establish NB cell line and cell suspension in our laboratory. Our experiments can supply a very important basis for the further research to the mechanism of disappearing spontaneously and the mechanism of inducing differentiation with gene and medicine.
