Objective: Study the role of dendritic cell in central nervous system on the course of experimental autoimmune encephalomyelitis (EAE) and tolerance induction by oral myelin basic protein.Methods: In order to establish EAE model, the mixture of MBP and CFA was injected to sjl mouse, with injecting bacteria of chincough in vein. To study the effect of oral tolerance, Some of those mouse took orally MBP before and day after injection. The time, incidence and course of each group were observed. At the same time, HE, LFB dyeing and immunochemist were used to study the infiltration of lymphocyte and dendritic cell in central nervous systemResults: No case occurred in control group. 75% of the mouse in EAE group developed the symptom and the course was 7.2±1.3.MBP prevent group and MBP treatment group came the symptom later(16.6±0.6day, 13.9±1.24), and had a lower clinical grade(4.6±0.4, 3.5±0.5). The Incidence of MBP prevent group (25%) are much lower than EAE group, while that of MBP treatment group had no obvious different between EAE group. There are few CD8a+ cell be detected in every group.Conclusions: 1. Dendritic cells are present at natural CNS and the CNS of EAE. Suggest it is type 1 dendritic cells other than type 2 dendritic cells play an important role in the pathology of self-immunity dieases. 2. Orally intaking MBP can ease the impairment of the EAE. 3. The dendritic cells in CNS of MBP treat animal are much fewer than untreated animal. This suggests the effect of dendritic cells in CNS immunity dieases.
