Objective To investigate the effects of all-trans retinoic acid(ATRA) on intimal proliferation , expression of proliferation cell nuclear antigen(PCNA),cyclin E, matrix metalloproteinase (MMPs) , and fibronectin (FN) in the Injured Rabbits carotid artery.to determine whether ATRA can prevent restenosis after coronary interventions therapy and the mechanisms.Methods 48 male New Zealand Rabbits were randomly allocated into four groups: normal diet group, control group,surgery group and ATRA treatment group. Normal diet group was fed by normal diet as blank control. Control group were fed by high cholesterol diet .Surgery group and ATRA treatment group were fed by high cholesterol diet and treated by air drying. ATRA treatment group were given ATRA(20mg.d-1,)into stomach immediatedly after injury . The carotid artery tissues were gathered calculated morphologically at day7, 28 after injury and the expression of PCNA, cyclin E,MMPs,and FN were measured by means of immunohistochemical technique and imagine analyzer.Results (1)The treatment of ATRA had no effect on serum lipids levels and body weight.(2) The typical carotid atherosclerosis lesion can induced by cholesterol diet plus air drying in rabbit. In surgery group(7d and 28d),there were distinct intima proliferation and luminal area decrease. (3)Histomorphometry revealed that there was no difference in luminal area(0.783 ±0.034 vs 0.829 ±0.039,p>0.05),neointima area(0.783 ±0.034 versus 0.829±0.039,p>0.05), medial area(0.544± 0.058 versus 0.540 ±0.07l,p>0.05)), intimal/medial area ratio(0.135±0.023 versus 0.128 ± 0.026,p>0.05),lumenal stenosis rate(2.334±0.221 versus 2.312±0.232,p>0.05) between surgery group and ATRA treatment group at day 7 after injury.However ATRA-mediated reduction was found in neointima area(0.337 ± 0.034 versus 0.181 ± 0.033,p<0.05), intimal/medial area ratio(38.72 ±5.113 versus 22.02 ±3.803,p<0.05),lumenal stenosis rate (56.83 ±13.24 versus 28.52±14.4l,p<0.01)between surgery group and ATRA treatment group at day 28 after injury. Luminal area was increased in ATRA treatment group (0.423 ±0.058 versus 0.556±0.067,p<0.05) at the same time.(4)Histomorphometry also revealed that the secretive quantity of collagen was greater at 28th day than 7 th day. compared with the surgery group , quantity of collagen of the ATRA treatment group were not decreased at 7th(11.56±6.33 versus 10.13 ± 7.11, p>0.05) and 28 th day(82.91 ±12.2 versus 52.34 ± 10.4,p<0.01) after injury. (5)Immunohistochmical analysis revealed that, there was no expression of PCNA, cyclin E, MMPs, and a little expression of FN in nomal group. (6) compared with the surgery group , expression of PCNA of the ATRA treatment group were significantly decreased at 7 th (42.7 ±8.82 versus20.7±7. 44,p<0.01)day...
