Therapeutic Effect Of Fluvastatin In Bleomycin-induced Pulmonary Fibrosis Of Rats

Abstract/Summary:
Objective To investigate the therapeutic effect of fluvastatin in bleomycin-induced pulmonary fibrosis of rats.Methods ① 45 male Wistar rats was divided into three groups: the normal control group(Group N), the bleomycin group(Group B)and the fluvastatin group(Group F). Group B received 5mg/kg of bleomycin intratracheally. Group F received the same amount of bleomycin as Group B, followed by 10mg/kg/d of fluvastatin. Group N received normal saline intratracheally. These rats were killed on 7,14,28 day post bleomycin. The lung tissues were obtained. ②Apply the HE staining to investigate the lung inflammation and lung fibrosis under the routine microscope. ③ Use the immunohistochemistry staining to study the expression of collagen Ⅰ and collagen Ⅲ in the lung tissues.④ Investigate the proliferation and apoptosis of the lung fibroblast with PCNA and TUNEL technique ,and calculate the proliferation index and the apoptosis index.⑤ The data were analysed by using logistic regression analysis and variance analysis.Results ①The degree of the lung inflammation and fibrosis were lower in Group F than in Group B(F/B 0R=4. 327>1 ; F/B 0R=5. 071>l).②The expression of Collagen Ⅰ and collagen Ⅲ in the lung tissue were less in Group F than in Group B(P<0.001). ③ The proliferation index of lung fibroblast was lower in Group F than in Group B(P<0.01; P<0.001;P<0.001). The apoptosis index of lung fibroblast was higher in Group F than in Group B(P<0.01; P<0.001;P<0.001).Conclusion Fluvastatin might be a therapeutic medicine for lung fibrosis.
