Object To observe pelvic-inflammation influence to endometriosis growth of early stage. To evaluate effects of anti-cyokines,danazole,and interferon on the growth of endometriosis lessions in the rat model. Method One hundred and five female Wistar rats were divided into two groups. Sexually mature female Wistar rats were used in this study. Fifty-five rats of artificial estrus were used in the experimental induction of endometriosis. The rest of female Wistar rats were injected 2% agal-agal in pelvic. Two weeks after the first surgery , endometriosis was induced surgically under anesthesia, acoording to procedures established. One month after the initial surgery, each rat was anesthetized and a midventral laparotomy was performed to determine the state of the endometrial explants. Each one was measured and photographed during each surgery. Fourty-eight endometriosis rats (EM rats) and fourty-five pelvic-inflammation and endometriosis rats (PID+ EM rats) were started on a treatment program. Fourty-eight EM rats and fourty-five PID+ EM rats were divived randomly into six groups and given the following agents for three weeks: danazole, anti-cyokines, antiauxin ,saline solution(control group). Three weeks after the initial treatment, laparotomy was performed and the explants were measured and photographed. The rats were killed by an overdose of anesthesia. Each transplant then was excised from the surrounding tissue, trimmed of fat, and embedded in paraffin wax for immunnohistochemical examination. We investigated TNF-α,IL-6 by immunnohistochemisity method . Result Pelvic-inflammation had no effect on the endometriosis rats of early stage .The length of explants in EM rats and PID+ EM rats was similar in both groups. No significant differences (P>0.05) in the tow groups .In addition to control group, the length of explants were decreased in five other treated groups. The anti-cyokines united with danazole group was found to be the most effective agents of the regression of endometriosis. In anti-cyokines group, the expression of TNF-α,IL-6 was decreased markedly. Conclution 1. Pelvic-inflammation hardly influence endometriosis growth of early stage, probably because of endometriosis. 2. Anti-cyokines caused regression of the explant in a rat model of endometriosis. It could reduce the length of explant and decrease the expression of TNF-α,IL-6. 3. The effect of anti-cyokines united with danazole group was greater than danazole or anti-cyokines group. 4. The effect of antiauxin is poor and antiauxin united anti-cyokines had no synergism. |
