Objective: The injury of the hepatic ischemia-reperfusion is always accompanied by intestinal endotoxemia(IETM).Both hepatic ischemi-reperfusion and intestinal endotoxemia stimulator aggravated the injury devived from producing considerable amount of pro-inflammatory cytokines and reactive oxygen species(ROS)of Kupffer cells.In the current study, we evaluated the effect of the administration of GdCl3 blockading Kupffer cells on the injury of ischemia reperfusion in the rat liver through operation of hepatic I/R,culturation of cells in vitro and administration of lipopolysaccharide(LPS). Methods: Experiment one: Male Wistar rats were randomly divided into three groups.Control group:Animals received a laparotomy without addition operation. I/R group:Animal of I/R group received an infusion of acidic 0.9%NaCl solution in a comparable volume for 24 and 48 hours before the operation of ischemia and reperfusion were made. GdCl3 group: Animals were pretreated with GdCl3 in a dosage of 10mg/kg (0.5%GdCl3 solution, pH 3.5). The operation was performed before GdCl3 was respectively infused into the tail vein for 24 and 48 hours.An atraumatic clip was placed across the portal vein, hepatic arrrtery, and bile duct just above the branch of the right lateral lobe .After ischemia for 45min, the peritoneal cavity was sutured shut, and animals were allowed to recover and to reperfuse for 3h and 6h.Following this period, serum and tissue were collected from each animal and frozen at -70℃ for subsequent liver enzyme and cytokine determinations,histopathological assessent,MDA,GSH and TNF-α content in the liver. Experiment two: Animals groups were the same as experiment one. Animals were used as KCs isolated to assess function of KCs's phagocytose, number of KCs's and ability of KCs's secretion after ischemia for 45min and reperfusion for 3h .Content of the TNF-α...
