| Objects: To set up the animal model of fulminant hepatic failure (FHF) inducedby thioacetamide TAA) in rats, to observe the change of cytokines on liverinjure and hepatic cells regeneration in each group and probe into the influenceof liver injure on hepatic cells regeneration in FHF.Methods: FHF was developed by subcutaneous injection of TAA. The rats wererandomly divided into 5 groups: the group of normal control(N),the group of 3hours(3h), the group of 6 hours(6h), the group of 12 hours(12h),the group of 24hours(24h). All rats were not killed till the end of the 3h, the 6h, the 12h and the24h by subcutaneous injection of TAA 500mg/kg for two days, at the same time.IETM was evaluated by plasma endotoxin; Liver function was observed by ALT,TB of plasma; The change of cytokines on liver injure and hepatic cellsregeneration will be demonstrate by TNF- α , IL-6, AFP, TGF- αimmunoradiometrical in the liver tissue; Observing the hepatic histopathologicchanges by HE staining and hepatic cells regeneration by proliferating cellmuclear (PCNA) immunnohistochemical staining.Results: The levels of plasma endotoxin, ALT and TB gradually decreased (vsN group, P<0.05). The content of TNF-α and AFP in the liver tissuesignificantly decreased (vs N group, P<0.05). The level of IL-6 and TGF-α inliver tissue have decreased (vs N group, P<0.05). The hepatic histophatholopicfeatures by HE staining showed the hepatocytes inflated, a large numberinflammatory cells infiltrated and point or slice necrosis in the liver tissue. Theimmunohistochemical staining showed that the PCNA of liver tissue expressionupregulation in FHF (vs N group,P<0.05).Conclusion: Intestinal endotoxemia (IETM) exit in the course of occurring FHF,which can induce the changes of cytokines. Such as proiflammatory cytokinesand liver regeneration cytokines. The degree of liver injure and the potentability to regeneration make a great impact on the development and prognosis ofFHF. |
