Objective: To study the expressions of gene and protein levels of metastasis-enhancing gene S100A4 and adhesion molecule CD44v6,E-cad,integrinαvβ3 in early stage squamous cervical carcinoma (SCC), explore their roles in lymphatic metastasis of SCC. Methods: Reverse transcription polymerase chain reaction (RT-PCR) was used to detect the mRNA expressions of CD44v6,E-cad and S100A4 in 21 normal cervival samples,23 SCC samples with lymph node metastasis and 63 SCC samples without lymph node metastasis. Immunohistochemistry was used to detect the protein expressions of CD44v6,E-cad,integrinαvβ3 and S100A4 in 21 normal cervival samples and 2 tissue chip(contain 63 SCC cases respectively) . Results: (1) The mRNA and protein expressions of CD44v6 were significantly higher and those of E-cad were significantly lower in SCC, a significant difference was both found between SCC with lymph node metastasis and without lymph node metastasis(P<0.05). The expressions of CD44v6 and E-cad were correlated with lymph node metastasis by Chi-square test(P<0.05).(2) The protein of integrinαvβ3 was weakly expressed in normal cervix and the extremely higher expression level of them were detected in SCC, a significant difference was found between SCC with lymph node metastasis and without lymph node metastasis(P<0.05)and the expression of integrinαvβ3 was correlated with lymph node metastasis(P<0.05).(3) The mRNA and protein expressions of S100A4 in SCC were significantly higher and lower in normal cervix. The expressions of S100A4 in SCC with lymph node metastasis were significantly higher than SCC without lymph node metastasis(P<0.01). Staining result of S100A4 was considered positive whenever increased cytoplasmic staining in SCC cells , fibroblast and lymphocyte. The expression of S100A4 in stroma cells was significantly higher than SCC cells (P<0.01),the expressions of S100A4 were correlated with lymph node metastasis by Chi-square test(P<0.05).The positive significant correlation is observed between the expressions of S100A4 and integrinαvβ3 by spearman rank correlation analysis. Though the correlation is not statistically significant between the expressions of S100A4 and CD44v6,E-cad,their correlation trend are observed. Conclusion: (1)CD44v6,integrinαvβ3,E-cad change cell adhesion of SCC cells to promot or suppress lymph node metastasis of SCC.(2)The expressions of S100A4 in SCC and stroma cells clue on its intracellular and extracellular functions and the induction of S100A4 expression leads to change cell adhesion of tumor cells and to an increased cell metastasis of SCC.(3)There are clinical significace in combining detection of the expressions S100A4 and CD44v6,integrinαvβ3,E-cad in SCC for predicting metastasis, evaluating prognosis.
