Objectives:To evaluate the expression of vascular endothelial growth factor-A, -C, R-3 mRNA of tissue and to study the relation among the expression of the three factors with cervical lymph nodes metastasis in laryngeal squamous cell carcinoma (LSCC) to investigate the mechanism of development and lymphatic metastasis. And compare the expression of tissue and those of cultured cell in vitro to provide the experiment base for the establishment of human laryngeal squamous cell carcinoma heterotrans-plantation model.Methods: VEGF-A, VEGF-C, VEGFR-3 mRNA in 24 cases of laryngeal carcinoma, 5 cases of normal mucosa epithelium around the cancer and 6 cases of cultured cell in vitro were detected by in situ hybridization. To study the relation of the three factors with clinicopathological parameters and lymphatic metastasis, the laryngeal carcinoma were divided by age, T stage, pathologic stage and lymphatic metastasis. Compare the expression of tissue and those of cultured cell in vitro.Results: Of the 24 tumors, In situ hybridization show the expression of the vascular endothelial growth factor-A, -C, R-3 mRNA was significantly higher in laryngeal carcinoma than normal mucosa epithelium around the cancer. Expression of VEGF-A mRNA was correlated with T stage, nor with age, pathologic stage. But expression of VEGF-C mRNA, VEGFR-3 mRNA was not associated with T stage, age, pathologic stage. The overexpression of VEGFR-3 mRNA had a positive correlation with the higher expression of VEGF-C mRNA. The overexpression of VEGF-A, VEGF-C, VEGFR-3 mRNA was related to cervical lymph nodes metastasis, and significantly statistical differences exist between the quantitative expression of three factors...
