Objective:The aim is to investigate the correlation of PARP with the metastasis of colorectal carcinoma , and how PARP may play its role. Methods: Immunohistochemical S-P was used to detect the expression of PAR ,P-selectin and ICAM-1 in colorectal carcinoma . The co-expression of PAR with P-selectin / ICAM-1 in colorectal carcinoma was investigated by immunofluorescence double staining. The effect of 5-AIQ on the adhesion of colorectal carcinoma cell line HT-29 to HUVEC was detected by adhesive experimentation .The immunocytochemistry and immunofluorescence double staining were used to investigate the expression of PAR ,P-selectin and ICAM-1, and the co-expression of PAR with P-selectin / ICAM-1 in HT-29 cell respectively. Results: 1. The expression of PAR, P-selectin and ICAM-1 was significant higher in colorectal carcinoma than that in the control colon mucosa (P<0.05) . Moreover , the expression of three proteins seemed to be closely related with the lymph node metastasis and(or) distant metastasis , but not with the sex , age , pathologic grade and the depth of infiltration . In addition , there is a positive correlation of PAR with P-selectin / ICAM-1 . 2. The adhesion HT-29 cell to HUVEC was reduced by 5-AIQ (P<0.05) . The expression of PAR, P-selectin and ICAM-1 was significant lower in 5-AIQ-treated HT-29 cell group than that in control group (P<0.05) . In addition , PAR co-expresses with P-selectin / ICAM-1 in HT-29 cell, too. Conclusion: These results suggest that the activity of PARP was increased in colorectal carcinoma, and the activity of PARP in colorectal carcinoma is closely related to the metastasis of colorectal carcinoma . PARP may play a role in upregulating the expression of P-selectin and ICAM-1 .
