Objective: To investigate the enhancement effect of the combination of wildtype p53 gene and Diacetyldianhydrogalactitol (DADAG/DADAD) on the growthand apoptosis of human hepatocarcinoma cell line HLE. Methods: The HLE cellswere transfected with plasmid pUHD10-3 recombined with wt-p53 gene bylipofectamine-mediated were cultured in medium containing DADAG.3-(4,5-Dimethy-2-thiazolyl)-2,5-diphenyl-2H-tetrazolium-bromid(MTT) techniquewas used to analyse the growth of HLE cells; and apoptosis of cells was evaluatedby flow cytometry and DNA electrophoresis. The expression of p53 was detectedby RT-PCR method. Results: The growth of HLE cells were inhibited after beingtransfected with wt-p53 gene, and 67.68% of the cells were inhibited on the fourthday. 48.17% of the HLE cells were inhibited when they were exposed to DADAG.When the HLE cells transfected with wt-p53 gene were exposed to DADAG,85.37% of the cells were inhibited at the 4th day. The apoptosis rates of the HLEcells after 96-hour treated were: 1.44% in untreated group, 3.53% inpUHD10-3-transfection group, 32.60% in DADAG group, 43.40% inpUHD10-3-p53-transfection group, and 74.60% in pUHD10-3-p53-transfectionplus DADAG-treatment group. DNA ladder was observed in pUHD10-3-p53-transfection plus DADAG-treatment group. The expression of p53 was low in HLEcells but high levels of p53 expression were observed in pUHD10-3-p53-transfected cells. Conclusion: The growth inhibition and apoptosis of HLE cellscould be induced by both wt-p53 gene and DADAG, and the effect was moreobvious when HLE cells were treated by the combination of wt-p53 gene andDADAG.
