Objective To study the effect of WWT extract on anti-inflammatory and analgesic and its anti-inflammation mechanisms. Methods (1)Croton oil-induced ear swelling, carrageenan-induced hind paw edema, the increase in celiac vascular permeability caused by acetic acid, the mouse pleurisy induced by carrageenan, and the weight of cotton granuloma in mice were prepared to study the effects of WWT extract on inflammation. The level of PGE2 in the paw was examined to explore its anti-inflammation mechanism primarily. (2)The residual content of Congo Red and carbon mote in mice blood were measured by the ultraviolet spectrophotometer to investigate the effect on nonspecific immune function. The effect on mouse spleen lymphocyte proliferation induced by Con A was tested by MTT assay. Serum pharmacological method of Chinese material medical was adopted too. (3)The bacteriostasis was conducted by testube and plate methods. (4)The hot plate test and acetic acid writhing method were adopted in the experiment to study the analgesic effect of WWT extract. (5)The coagulation time was measure by turbidimetry. (6)Qualitative analysis of WWT extract were also tested . Results The ear edema induced by Croton oil,paw edema induced by carrageenan,the increase in vascular permeability caused by acetic acid and weight cotton granuloma in mice were inhibited significantly by WWT extract, it also inhibited the chemotaxis.WWT inhibited the synthesis of PGE2 in the injected paw of rats caused by carrageenan. WWT extract can strengthen the function of engulfing Congo Red and enhanced the index of phagocytosis of carbon mote in mice.It also stimulated the proliferation of mouse spleen lymphocyte induced by Con A. In the hot plate test, after inject WWT, antinociceptive action was present with the pain threshold improved and showed strong periphery analgesic action, it had been proved that WWT provided good analgesia in writhing induced by acetic. WWT extract significantly inhibited the growth of bacillus coli, staphylococcus aureus,the MIC was 0.002,0.032g/ml,respectively. It also shortened the time of homeostasia but elongated the time of blood coagulation.The determination of flavona,saponin, organic acid, carbohydrate, alkaloid, phenols and tannin, phytosterols, triterpenoids were positive findings. Conclusions WWT extract has anti-inflammatory and analgesic effects. The anti-inflammation mechanism may be related to strengthen the phagocytes of macrophage, promote T lymphocyte proliferation and decrease PGE2 level.
