Peripheral nerve exists in the body extensively,it is ofteninjuried because of trauma and compression .Plerosis andregeneration of peripheral nerve after injuried is always animportant research subject in trauma surgery .The local injuryfactors are important in the past about the factors affectingrecovery of function after peripheral nerve injury,but the studyof neuron is not important. In fact, as a cell entirety,the neurongets in touch with axon intently, the injury of nerve fibre willinvolve with the neuron certainly,and the function status of neuronalso affect the recovery of peripheral functionpost-injury.Therefor ,the study of neuron following peripheralnerve injury has become the hot topic in the peripheral nerve field .Some studies investigate that the peripheral nerve injury willresult in a certainly number neuron degeneration and death,andneuron nissl body will collect or lysis , cell nucleus swell ,sidemoving and disintegrate,and cell plasm dissolve ,even die. Aboutthe effect of post-peripheral nerve injury to neuron ,the reportresults are very much ,the degree is not identical, but there aretwo effect factors to be common,it is that the death number of neuronand the age is inverse ratio,and the degree and distance of injuryis inverse ratio .At the same time ,post-peripheral nerve injurywill inevitably result to a certainly number neuron death, distalnerve degeneration ,myelin sheath and axon degeneration andnecrosis .Because neuron segregates with terminal organs, nerveterminal and effective appararus also become atrophy anddegeneration . Post-injury peripheral nerve has strongregeneration ability ,how to accelerate peripheral nerveregeneration earlier and better and recovery normal nerve functionis the importance and difficulty . Many medicines can acceleratethe recovery of center nerve , those include nerve growth factorand neurotrophic factors and so on .Because of expensive price andside effect , these use in clinic uncommonly .Recently ,many distillcomponent of Chinese traditional medicine is used in nerve injuryrecovery ,according to experimental study ,both monomer componentand compound component have certainly effect to peripheral nerveregeneration .In order to discuss the effect of Chinese traditionalmedicine to protect neuron and accelerate peripheral nerveregeneration ,we devised the experiment as follow .Objective To investigate the protection and regulation effectof compound Chinese herbs to nerve regeneration and neurons ofspinal cord anterior horn following peripheral nerve injury inrats.Method: Forty-two adult Wistar rats weighing about 200g wererandomly divided into two groups(experimental group and contrastgroup).The rats were sacrificed on postoperative week4,6,8respectively to detect motor induced potential conduction velocityand amplitude of sciatic nerve by electromyography;The number ofmyelinated nerve axon and cross section area of myelinated nervefiber were detected by histology .The morphologic changes and theultrastructural changes through scanning electron microscope.HEstaining of spinal cord anterior horn was used to investigate theprotection and regulation effect of compound Chinese herbs to nerveregeneration and neurons of spinal cord anterior horn followingnerve injury .Results :It was found that nerve conduction velocity,regeneratedmyelinated nerve fiber cross section area and number of nerve fiberwere superior to those of the contrast groups ,and the neuronaldegeneration level in spinal cord anterior was less in theexperimental groups than in the contrast groups ,the member ofsurvial neurons were significantly higher in the experimentalgroups than in the contrast groups . At fourth and sixth week,it was found that nerve conductionvelocity and amplitude of experimental group were superior tocontrast group ,regenerated myelinated nerve fiber cross sectionarea and number of nerve fiber were superior to those of the contrastgroup among HE staining specimens. At sixth and eighth week,according to the electron microscope results,in exeperimentalgroup,there is no interspace between neuron andneuropil ,karyotheca is regular ,there are much euchromatin androuth endoplasmic reticulum and mitochondria in nucleus . Partmitochondria swell lightly and degree of vacuole little. There islarger interspace between neuron and neuropil ,neuron is darkcell ,nucleus brim is not regular , Stroma is dense ,neurite becomevacuole shape ,neurofilament reduce,mitochondria swellclearly,and most mitochondrial crisrae rupture.
