Objective To investigate the changes of intra-arterial activity ofextracellular regulated protein kinase (ERK) and cyclooxygenase-2(COX-2) in atherosclerosic rabbits and further evaluate the effects ofangiotensin converting enzyme inhibitor(ACEI) captopril on this pathway .Methods Japanese flap-eared rabbits were randomly assigned to threegroups : non-atherosclerosic control group , non-treated atherosclerosicgroup and captopril-treated atherosclerosic group . Non-treatedatherosclerosic group and captopril-treated atherosclerosic group weregiven cholesterol diet twelve weeks, Two rabbits of each CH-fed grouphave be stochastic draw-out and to observed whether or not success ofatherosclerosic models . and then , the captopril-treated atherosclerosicgroup were given captopril , Four weeks later the expression ofcycloooxygenase-2(COX-2) and phosphatic extracellular regulatedprotein(p-ERK) in atherosclerosic plques were inverstigated onpathological sections by monoclonal antibody immunohistochemistrystaining and the serum cholesterol (CH) , triglycerides (TG) , high-densitylipoprotein(HDL) and low-density lipoprotein(LDL) and thehistopathological changes of aorta were detected in the various groups .Statistical analysis: Numerical variable datas were expressed by X±s. Thedatas compared between two groups used analysis of t-test, more thantwo groups the analysis of variance and q-test were used to comparedifferences by SPSS12.0 statistical software. P<0.05 that meant significantdifference. Results 1,The fatty streak were obviously in the two CH-fedanimals and the layer of foamcells were remarkably increasing after twelveweeks . The atherosclerosic models were succeed . 2,The serum CH , TGand LDL in non-treated atherosclerosic group and captopril-treatedatherosclerosic group increased compared to the non-atherosclerosiccontrol group and compared to the non-treated atherosclerosic group , theserum CH , TG and LDL were not decreased in the captopril-treatedatherosclerosic group after sixteen weeks . 3,The aortio neointima wassmooth and glossy in the non-atherosclerosic control group , when thenon-treated atherosclerosic group and the captopril-treatedatherosclerosic group's fatty streak were obviously and the layer offoamcells were remarkably increasing after sixteen weeks and comparedto the non-treated atherosclerosic group , the layer of foamcells wereremarkably decreasing in the captopril-treated atherosclerosic group . Thelarger ratio of aorta plaque areas,the larger ratio of aortio neointima areasand medius-membranous areas and the smaller lumen areas innon-treated atherosclerosic group and captopril-treated atherosclerosicgroup increased compared to the non-atherosclerosic control group andcompared to the non-treated atherosclerosic group , the smaller ratio ofaorta plaque areas,the smaller ratio of aortio neointima areas andmedius-membranous areas and the larger lumen areas were proved incaptopril-treated atherosclerosic group . 4,Augmented expressions ofCOX-2 were demonstrated in the atherosclerosic plaques , few expressionof COX-2 in the vessel smooth muscle below the plaque in the two groupsof CH-fed animals and compared to the non-treated atherosclerosic group ,the COX-2 were decreased in the captopril-treated atherosclerosic group...
