Objective To observe the level of expression of CD25 on T lymphocyte surface in congestive heart failure;to explore the effect of carvedilol on expression of CD25 of human T lymphocyte in vitro; to investigate the effect of carvedilol on in vitro mononuclear cells TNF- α and IL-6 expression and production in human peripheral blood.Methods (1) Patients with chronic heart failure have a LVEF less than 0.50,as experimental group (n=18),control groups are healthy volunteers(n=12).The expression of T lymphocytes activation markers such as CD25 and CD3 on cell surface in two groups was assayed with flow cytometry in order to investigate the levels of expression of CD25 on T lymphocyte surface in patients with chronic heart failure and normal donors, and the difference in two groups ; (2)peripheral blood mononuclear cells(PBMC) were prepared from normal donors, and incubation with phorbol 12-myristole 13-acctate(PMA) and A23187 with or without carvedilol in vitro .After incubation, The viability of PBMC was detected by MTT; the early and late apoptosis of PBMC was assayed with flow cytometry ,in order to definite the safety confine of carvedilol in vitro;(3)PBMC was isolated from healthy donors and incubated with PMA and A23187 with or without different concentration carvedilol in vitro ,carvedilol was added 2 hours before PMA and A23187 ,then incubated.After incubation 24 hours,the cells were collected and the expression of T lymphocyte markers such as CD25 and CD3 on cell surface was assayed with flow cytometry, in order to investigate the effect of carvedilol on CD25 expression of T lymphocyte surface in vitro; (4)PBMC was isolated from healthy donors and incubated in eight groups (see Figure 1) .
