Objective The experiment was carried out to study the effects of EGF and HFF in culture medium on in vitro maturation.fertilization and embryo developmental competence of human immatured oocytes.Mothods Human immatured oocytes came from patients with PCOS who were received IVM treatment.The patients were arranged randomlv in three groups.Control group:TCM199 +10 % SSS +0.075IU/mlFSH +0.5IU/mlHCG +1μg/ml17β-E2 +0.05mg/ml penicillinG +0.075mg/mlstreptomycin sulfate +0.2mM/L pyruvic acid; HFF group: control group medium+10 % HFF; EGF group: control group medium-r2ng/ml EGF. After 24-48h cultured, oocytes extruded the first polar body were inseminated by intracytoplasmic sperm injection(ICSI). 16-18h after ICSI, fertilization of oocytes were assessed and fertilized oocytes continued to be cultured 2-3 days. The maturation, fertilization, oocyte cleaved. 2-cell. 8-cell, high quality embryo, implantation and clinical pregnancy were observed.Result1 The effects of HFF and EGF on maturation.fertilization and cleavage rate on in vitro maturation of immatured oocytes. Cleavage rate in HFF group is increased significantly comparing to control group(P<0.01).but maturation.fertilization rate in this group have no statistic significance comparing to control group(P>0.05).MII rate,fertilizatin rate in EGF group are increased significantly(P<0.01).but there is no statistic significance of cleavage rate in EGF group comparing to control group(P>0.05).2 The effects of HFF and EGF in culture medium on embryo developmental competence of IVM. High quality embryo rate in HFF group is increased significantly (P<0.05), but other rates in this group have no statistic significance as compared to control group(P>0.05). 8-cell rate and high quality embryo rate in EGF group are increased significantly(P<0.05). but there is no statistic significance of 2-cell rate in EGF group as compared to control group(P>0.05).3 The effects of HFF and EGF in culture medium on implantation rate and clinical pregnancy of IVM. Implantation rate and clinical pregnancy rate in HFF group are increased significantly comparing to control group(P<0.05), but there are no statistic significance of implantation rate and clinical pregnancy rate in EGF group as compared to control group(P>0.05).Conclusion1 HFF can improve cleavage,high quality embryo and clinical pregnancy rate in human immatured oocytes, which suggests that HFF can enhance cytoplast maturation of oocytes in vitro.2 EGF can improve maturation.fertilization.eight-cell and high quality embryo rate in human immatured oocytes, which suggests that EGF can enhance nuclear maturation and cytoplast maturation of immatured oocytes in vitro.
