Objective To study the therapeutic effects and mechanisms of SQDG on carbon tetrachloride -induced chemical liver injury in mice , hepatic fibrosis in rats and primary cultured rat hepatocytes as well as its possible mechanisms . At the same time the pharmacodynamics of SQDG was compared with total glucosides of paeony (TGP) or astragalosides (ASTs) of effective dose.Methods The model of carbon tetrachloride -induced chemical liver injury in mice, hepatic fibrosis in rats and primary cultured rat hepatocytes was prepared. The levels of alanine aminotransferase(ALT), aspartate aminotransferase(AST) , nitric oxide(NO) in serum, hydroxyproline content, malondiadehyde(MDA) content, superoxide dismutase(SOD) and glutathione peroxidase(GSH-Px) activities in liver homogenate were assayed by spectrophotometry; serum hyaluronic acid (HA), laminin (LN) procollagen III (PCIII), collagen type IV (CIV) and TNF-a activity were determined by ridioimmunoassay. The survival rate of CCI4- injury hepatic cell was assayed by MTT dye reduction. Meanwhile, hepatic pathological examination was observed.
