ObjectiveTo evaluate the results of bone marrow mesenchymal stem cells (MSCs) transplantation combination with recombinant human Growth Hormone(rhGH) in rats with acute myocardial infarction.MethodMSCs cells were collected from bone marrow of Wistar rat's femoral and tibial bones,cultured and passaged in vitro,then purified by adhesive-screening method.The animal with acute myocardial infarction were built by direct liquid nitrogen cryoinjury with a metal probe.Thirty-eight Wistar rats were randomly divided into 5 groups: sham operation group(S group,n=6), MSCs transplanted group(M group,n=8), NS injection group(C group,control group,n=8), rhGH treatment group(G group,n=8), MSCs combined with rhGH group(MG group,n=8).In the M and MG group, the MSCs(12.5 ×106cells/50μL) labelled with BrdU were directly injected into the cryoinjuried area. The next day, rhGH were given subcutaneously 2IU ? kg-1 ? d-1 × 14d in group G and MG At the end of 4 weeks, the rats were euthanized, their heart weight and body weight were measured, the ratio of body and heart weights calculated. The density of capillary were counted. The differentiation of MSCs was evaluated by immunohistochemical double stain of BrdU and cTnI(cardiac troponin I).ResultThe cultured MSCs were spindle-shaped, attached to the culture dish tightly,the purified MSCs were acquired on the day 14. The cryoinjury pathogenesis included necrosis, inflammatory reaction of infarcted area, ventricular remodeling of LV was similar with the pathologic changes of humanity. At the end of 4 weeks , the body weight,heart weight of the rats from group G and MG were higher than those in the other groups, but rhGH treatment did not cause significant differences in the ratio of heart weight to body weight(P>0.05). No difference in infarcted sizes was found among all groups. The average infarced size was about 20.36%. The density of capillary(the number of capillary/0.2mm2 )was higher in group M(7.44±1.85), G(7.25±2.28),and MG(9.30±2.24) than that of group C(3.60±1.39, P<0.05). The density of capillary in group MG was much higher than those in G and M group respectively (t= 1.8571, 2.0500, P<0.05), the MSCs and rhGH had similar effect on angiogenesis. BrdU-positive cells were observed at infarcted area in groups M and MG with MSCs transplantation, there were no BrdU-positive cells in infracted area of the control group, the number of BrdU-positive cells in group MG was much more than those of the group M. BrdU-stained cells were also exist in endothelium. Cardiac-like cells which stained positively for both BrdU and cTnl were observed at the edge of the infarcted scar area, especially in MSCs transplantation combined with rhGH group.ConsultionBone marrow-derived mesenchymal stem cells could be obtained by using density gradient centrifugation and adhesive culture methods. RhGH could increase heart weight and body weight, but did not change their ratio. Both MSCs and GH could increase the capillary density of infracted area significantly.
