Human breast carcinoma is one of the most frequent cancer in the female malignacies and deeply threatens woman health and life quality.Because of its carcinogensis and development with lots of genes alteration, it becomes a very important region in searching the novel effective gene and studying its mechanism in breast cancer evolution.ANGPTL4 is a novel angiopoietin-relatived gene, previously named as pp1158 and now it has been named as ANGPTL4 by HUGO. It contains integral 3' terminal and 5' terminal, and the initial translational region with the Kozak context. The full length cDNA of ANGPTL4 consists of 1943 nucleotides in which there is a 1218 bp and ORF encoding a 406-amino acid polypeptide,predicted molecular mass is 45.2kDa. A conserved coiled-coil domain and a signal peptide were found to be present in the NH2-terminal region and a fibrinogen-like domain in the COOH-terminal region and a potential N-glycosylated site and a few phosphorylation sites. ANGPTL4 share 40% amino acid identity with Ang2. ANGPTL4 is a secret protein and is mapped on chromosome 19pl3.3, which is the most frequently mutated region on chromosome in human cancers.We discuss the relationship between ANGPTL4 and human breast carcinogenesis.
