Background: The metastasis and recrudescence is the important reason with the breast cancer that cause the patient death, and is the difficulty of clinical therapy in breast cancer. It is a very complicated and a sequence process that breast cancer metastasis and is numerous factors to moderate mutually and coefficient result.This laboratory screened a gene-KNSL4(kinesin-like DNA binding protein )that expressed is differential in primary breast cancer with its transitivity cancer use of the gene chip technique, mRNA differential display technique. Our researche adoption real-time Q-PCR method to verify the relativity of the expresses level of KNSL4 gene and the metastasis of breast cancer , And applied small interference RNA( siRNA) technique to verify the influence to the ability of metastasis to breast cancer MDA- MB-453 cells of KNSL4 gene, certain the function of KNSL4 gene in breast cancer transfer and prognosis, evaluate its meaning in mammary glands cancer study.Objectives: Study the relativity of kinesin-like DNA binding protein (KNSL4) with metastasis of breast cancer.Method: 1. Adopt real-time Q-PCR method examine KNSL4 genes in the level expression in mRNA of 108 clinical specimens and 9 breast cancer cell lines, analyzing its relation with metastasis of breast cancer and the the clinical pathologic index.2. Adopt siRNA technique proceeds the interference to the high expression in KNS1A MDA- MB-453 cell inside KNSI4 gene, and apply the Matrigel invasion Chamber examination the siRNA interference in front and back the MDA-MB-453 cells the changes of the ability of metastasis o Results:1.
