Objective: To study the effects of Artesunate on immuno-injured hepatic fibrosis induced by bovine serum albumin in rat model. And to investigate the influence of Artesunate on proliferation and cell cycle of hepatic stellate cell line-T6 and explore its mechanisms of antifibrosis.Method: Experiment 1. The model of immuno-injured hepatic fibrosis induced by bovine serum albumin was established in Wistar female rats. Rats were randomly divided into three groups, namely a normal control group, a model group, an interventional group (3.75 mg/200 g). Except for normal group, rats in other groups were given bovine serum albumin to induce hepatic fibrosis. Rats in all groups were given corresponding drugs .At the end of week 15, all the rats were sacrificed. The pathological changes of liver were observed by HE and Masson staining under microscope. The content of hydroxyproline of liver tissue, serum liver function, MDA were analyzed by colorimetry.Experiment 2. The rat hepatic stellate cell line, HSC-T6 was generationally cultured in DMEM medium. HSC-T6 was incubated with different concentration of Artesunate. The inhibition of cell proliferation was determined by MTT method. Cell cyle was determined by flow cytometry analysis. Apoptosis was confirmed by ladder DNA electrophoresis. Cytotoxicity was measure by LDH colorimetry. The content of hyroxyproline was measured by colorimetric method.
