Objectives To detect the expression and distribution of bcl-2 and bax in the nasal polyps tissue and explore the induction and control effect of bcl-2 and bax in the pathogenesis of human nasal polyps.Methods With the methods of the SABC technique ofimmunohistochemistry, bcl-2 and bax gene expression were measured between 30 human nasal polyps tissues and 30 inferior turbinate mucosas tissues, eosinophieis was identified with HE staining. The appearance of brown-yellow pigmentation in cytoplasm was considerd as positive cell. Microscope was used to calculate the pigmentation of each segment subjectively and ascertain the positive cell expressive rate. The result expressed as norm ± standard deviation, all the results were disposed with statistical analysis.Results The results indicated that (1)there was a significant overexpression of bcl-2 on nasal polyps (epitheliums:82.47±7.16, glands:80.52±7. 02)compared to inferior turbinate mucosas(epitheliums:14.41±3. 30, glands:12.63±3.11) ,(t=12.22, t=12.50 ,p< 0.01); bax was overexpressed on epitheliums in nasal polyps(71.52±5.24) compared to inferior turbinate mucosas (9.36±2. 35),(9.36+2.35),(t=15.31 ,p<0.01),while glands, there are no significant overexpression(t=0.46,p>0.05);the expression of bcl-2 was significantly stronger than bax in glands of nasal polyps tissues(t= 14.31 ,p<0.01). (2) In eosinophieis,the positive cell expressive rate of bcl-2 was significant higher in nasal polyps than that in inferior turbinate mucosas(t=10.18 , p<0.01),while,the positive cell expressive rate of bax was slightly higher in nasal polyps than that in inferior turbinate mucosas(t=1.23,p>0.05).Conclusions There was significant difference of the expression of bcl-2 between normal inferior turbinate mucosas and nasal polyps in epitheliums, glands and eosinophieis,while the significant difference of the expression of bax was only in epitheliums.The study demonstrated that there were correlations between the occurrence and evolution of nasal polyps and the loss of bax and the raise of the expression of bcl-2.The high expressions of bcl-2 and low expressions of bax in nasal polyps tissues owed to the nasal polyp epithelial cell's escaping from cellular apoptosis induced by bax/bcl-2 system and excess mullcplication.Growing upwards led up to expanding of the volume of nasal polyp,and growing downwards led up to the formation of new glands in interstitial tissue.The apoptosis inhibiting of eosinophieis made it infiltrate in nasal polyp tissue.So we presumed that bcl-2/bax may play an important role on the pathogenesis of human nasal polyps, including cystic degeneration of submucosal gland, apoptosis inhibiting of eosinophieis.
