BACKGROUD: Animal ischemia model is the foundation in many reseaches, which includes neovascularization, the etiopathogenesis and therapy of ischemic disease or other contents. And hindlimb ischemia model is the widely used one of them, for it involved in the study of angiosclerosis and embolism in lower limb. Recentely, it becames a original tendency that encouraging the neovascularization by stem cell transplatation in animal hindlimb ischemia model. In the involved empirical studys, It is very important that triumphantly established a hindlimb ischemia models which have certain effect and high repeatability, for it is the key point of make the experiment proceeding smoothly. But after reading the interralated papers, we found that no reseachers have reported detailed studies involved hindlimb ischemia models. Although some reseachers mentioned the models in their papers, they describe the methods and results of hindlimb ischemia models very little.Furthermore, it is very defferent of the reseachers' expressions,and there is no unanimous conclusions.So, in this papers, we try to investigate the effect and repeatability of hindlimb ischemia models in details. First, We establish the hindlimb ischemia model in Wistar rat by ligating defferent arteries and observe its effect; Second, we reseach the vascular anastomosis in wistar rats' hind limb by ligating arteries andarterial perfusion. Besides discussing the disadvantages of ligating arteries, We establish Wistar rat hindlimb ischemia model by common bletilla tuber embolizing femoral artery, and we think it is a reliable and effective methods.PART 1:THE EFFECT OF WISTAR RAT HINDLIMB ISCHEMIA MODEL AFTER LIGATING ARTERIESOBJECTIVE: We establish the hindlimb ischemia model in Wistar rat by ligating defferent arteries and observe its effect.METHODS: We separate and sever defferent arteries of rats' hindlimb, and reseach if the ischemia appearing by pathology.RESULTS: There is no obvious changes in rat's hindlimb before and after ligation.CONCLUSION:1. There is no obvious ischemic changes after respectively ligating superficial femoral artery, femoral artery, external iliac artery and common iliac artery;2. Wistar rats have two blood-supply resources, internal iliac artery system and external iliac artery- femoral artery system. It can be the one of ligation fail's reasons.3. Because it can not explain the appearance of ligating common iliac artery, we think there can be other influencing factors.PART 2:THE TRACER STUDY OF VASCULAR ANASTOMOSIS IN WISTAR RATS' HIND LIMBOBJECTIVE: we reseach the vascular anastomosis in wistar rats'hind limb by ligating arteries and arterial perfusion.METHODS: Injecting Prussian blue-chloroform solution into aorta and reseaching the vascular display of wistar rats' hind limb. There is three group: control group,perfusing after ligating femoral artery, perfusing after ligating common iliac artery for three days.RESULTS: After ligating arteries, Prussian blue-chloroform solution still can circulate in the blood vessel of wistar rats' hind limb.CONCLUSION:1. There is affluent vascular anastomosis in wistar rats' double hind limbs and different location of each hind limb;2. Wistar rats' hind limb can still obtain blood supply after ligating arteries;3.Ligating rat's arteries does not cause hindlimb ischemia. PART 3:COMMON BLETILLA TUBER EMBOLIZING FEMORAL ARTERY ESTABLISH WISTAR RAT HINDLIMB ISCHEMIA MODELOBJECTIVE: We establish Wistar rat hindlimb ischemia model by common bletilla tuber embolizing femoral artery and invertigate its feasibility and effect.METHODS: After embolizing rat's femoral artery using common bletilla buber and other vascular occlusive agent (e.g.iodinated oil), we investigate the changes of hindlimb by pathology and other means.RESULTS: Comparing with other vascular occlusive agent, thereis much more obvious ischemia in rat's hindlimb after using common bletilla tuber superfine embolized femoral artery. CONCLUSION:1. After using common bletilla tuber superfine embolized femoral artery, severe chronicity continuity ischemia appears in the wistar rats' hind limb;2. Comparing with other vascular occlusive agent (e.g.iodinated oil), if using common bletilla tuber superfine embolized femoral artery, its ischemia effect is more obvious, ischemia duration is longer and reablement is more slowly;3.For hindlimb ischemia model, common bletilla tuber embolizing femoral artery is a ideal methods compared with other general ones.
