Objective: Identify the human embryonic germ cells by observing the expression of its specific surface markers SSEA-1,SSEA-3. Methods: Human embryonic genital ridges, dorsal mesenteries(5-9 weeks post-fertilization) were cultured without any cytokines, while the fibroblasts derived from the isogenous embryo were used as the feeder layer. We detected the expression of SSEA-1 ,SSEA-3 on the cultivated cells which similar to the embryonic germ cells,through immunohistochemistry;Paraffin sections were prepared from the the embryonic genital ridges of 5-9 weeks post-fertilization to show the PGCs,by observing the expression of SSEA-1 ,SSEA-3 on the PGCs, through immunocytochemistry. Results: Abundant PGCs with positive expression of SSEA-1 and SSEA-3,arranged in irregular cords,were found in the embryonic genital ridges of 5-9 weeks post-fertilization.While cultured in vitro,both the single cell and the cluster of cells which grew on the embryo fibroblasts showed the positive expression of SSEA-1,SSEA-3. Conclusion: Cells cultured in such a cultivate system came from PGCs,and definitely were undifferentiated human embryonic germ cells.
