Marine sulfated polysaccharide drug 911 is derived from brown algae, with good anti-virus activity.The purpose of this paper is to establish a microanalysis method of marine sulfated polysaccharide drug 911 in biological sample—rabbit serum, and to afford prophase study in demic pharmacokinetics. Using post-column fluorescent derivation HPLC to analyze 911 in rabbit serum, using guanidine hydrochloride as derivative reagent and ion exclusion column Aminex HPX-87H as analytical column.In the pretreatment of rabbit serum, contrasting with tri- chloroacetic acid, perchlric acid, acetonitrile, methanol, alcohol and saturated ammonium sulfate, enzyme-Sevag method is selected to remove protein. Design of orthogonal experiment to find the best experimental condition of enzyme. High performance gel permeation chromatography (HPGPC) is used to test the stability of 911 to pronase E through determing molecular weight and molecular weight distribution. The results show that 911 is stable to pronase E under this experimental condition.In this paper, post-column fluorescent derivation HPLC is firstly used to analyze 911 in rabbit serum. Building standard curve and liner range, the limit of detection and limit of quantitation are determined, that is 2.5μg·mL-1 and 10μg·mL-1, respectively. The precision reproducibility and recovery of 911 in rabbit serum are tested. This method is used to detect 911 in rabbit serum after ig. Experiments prove this method has high sensitivity, the precision and reproducibility are better. All these research provide basic method for 911 preclinicle and clinicle pharmacokinetics studies.
