Objective: The aims of the research is to find the possible uses oftrichosanthin(TCS) in the transplantation immunology ,through the prolongation of the survivals of the allograft in the skin transplantation model to establish the understandings of the mechanism of the drug in the cellular and molecular effects to the immune system, and the possibilities for TCS to be used as a immuno-suppressor in the organ transplantation ,to exploited the new use of this type I ribosome inactivation protein (RIP),may deepen the understandings in the transplantation.Methods: In this study allogeneic skin transplantation mice models are usedto observe the function of trichosanthin to the skin survival, and regulatory T cells (Tregs) and cytokines are tested in vivo in the peripheral blood by the Flow Cytometry and Luminex, the suppress of the ConA-stimulated lymphocyte proliferation are detected by ~3H-TdR incorporation. The changes of the expression of the ConA-stimulated lymphocyte surface molecular—activation molecular-CD69 are also be measured by Flow Cytometry. All these measures are used to help us to understandthe mechanisms of the drug to the immune system.Results: TCS prolonged the survival of the graft in the allogeneic skintransplantation in mice model, TCS can also suppress the proliferation of the peripheral lymph node lymphocyte which are stimulated by the multiple clone stimuli-ConA in vitro. The changes of the regulatory T cells and cytokine profile in the peripheral blood may contributed to the understandings of the cell and molecular immuno-mechanism for the phenomenon in the mice model. But the doubts still existed in the detection of the expression of the surface molecular of lympho node lymphocyte -CD69- stimulated by ConA in vitro -which was considered as an early activation marker of lymphocyte proliferation. No correlation are tested upon the use of TCS .Conclusion: Certain dose of TCS can suppress the immune response of thealloantigen in the mice skin transplantation model and maintain a certain degree of tolerance, maybe through the cytokine profile and (or) regulatory T cells (Tregs) mechanisms. Optimal protocols of the drug may be considered to reduce the side-effects of the normal suppressor such as CsA (cyclosporin A). Understanding of the functions and the immuno-mechanism of the trichosanthin to the immune system furthered, database could be considered to be prepared for the further expected use of the drug in the organ transplantation .
