Objective:To investigate the curative effects of homograft of mesenchymalstemcells(MSCs) complexed with medical collagen membrane of GTR (MCMG) onthe full thickness defects of articular cartilage.Methods: 1.The bone marrow wasaspirated from the bones of spina iliace of New Zeal rabbits and was isolated bygradient centrifugation. Flow cytometric analyses were performed to identify themononuclear cell. The higher purity of MSCs were obtained by the repeated removalof nonadherent cells .Observed the changes of MSCs morphology, growth andproliferation in vitro. The third passage MSCs labeling by hoechst-33342 fluorescentwere seeded onto medical collagen membrane of GTR (MCMG). The MSC/MCMGcomplex was cultured and transplanted into full thickness defects on medicalcondyle and trochlea of both knee jionts. 2.Twenty-seven healthy New Zealand rabbitswere divided into three groups randomly with 9 in each group. The cartilage defectswere transplantde with homograft of mesenchymal stem cells(MSCs) and medicalcollagen membrane of GTR complex (MSCs/ MCMG) in group A (management A) ,with only medical collagen membrane of GTR in group B (management B) and withnothing in group C (management C) . Three rabbits were killed at 4 , 8 and 12 weeksafter the operation in each group , and the reparative tissue samples were evaluatedgrossly ,histologically , immunohistochemically and graded by gross and histologicalscale .SPSS 11. 0 software was applied to proceed the statistic alanalysis.3.The sourceof the reparative tissue were detected . Results :1.It is a simple,effective and practicalmethod to separate and to obtain higher purity and activity of MSCs from the bonemarrow of rabbits by gradient centrifugation .MSCs can grow quickly when culturedin vitro in low abundance. 2.The defects in MSCs/ MCMG transplantation group wererepaired by hyline-like tissue and the other groups were repaired by fibrous tissues.Statistical analysis of gross and histological grading scales showed that the repairs inMSCs/ MCMG transplanta on medical condyle and trochlea are no different( P > 0.05). There is no significant difference between medical condyle and trochlea. MSCs/MCMG group was significantly better than MCMG group and control group ( P < 0.05) , and MCMG management was better than control management ( P < 0.05) .3.The source of the reparative tissue were transplanted MSCs . Conclusion:1.Transplantation of mesenchymal stem cells complexed with medical collagenmembrane of GTR (MCMG) on the full thickness defects of articular cartilage is apromising way for the treatment of cartilage defects. 2.The source of the reparativetissue were transplanted MSCs .
