Study On The Protective Effect Of Fu Fang Huang Jing Oral Liquid On The Myocardium In Rats With Adriamycin-induced Heart Failure

ObjectiveTo explore the protective effects of Fu Fang Huang Jing Oral Liquid onthe myocardium in rats with Adriamycin-induecd heart failure.MethodsFifty-five male SD Rats, weighed 190~210 kg, were randomly assigned intofive groups: the control group(SD); Group Adriamycin (ADR); Group Fu FangHuang Jing oral liquid at a small dosage (HJl); Group Fu Fang Huang Jingoral liquid at a moderate dosage(HJm);Group Fu Fang Huang Jing oral liquidat a large dosage(HJh). The control group have received peritonealinjection with normal sodium once a week ,for six weeks. Adriamycin wasgiven to the second to fifth groups with the dosage per 3mg/kg dissolvedin 1ml normal sodium once a week,all for six times.At the meantime Fu FangHuang Jing Oral Liquid was given to the third to fifth groups of rats for6 weeks,once a day,with the dosage by turns 2ml/d,4ml/d,6ml/d.In the courseof experiment ,observing the general state of health of the rats regularlysuch as the mental state,action,collar pattern,foodintake,Weight andascite.At the same time left ventricular hypertrophy index (LVHI) weremeasured. Angiotensin â…¡(Angâ…¡) and aldosterone (Ald) were detected byradio-immunity.Blood plasma collagen fibers were detected byradio-immunity . Ventricular fibrosis in ADR were evaluated according tocollagen volume fraction (CVF) and perivasular circumferentiall area(PVCA).The constitute of myosin light chain 2 were measured by weasternblotting. Ultrastructural features of left ventricular were observed bytransmission electron microscope (ECM).Results 1.After six-weeks treatment, the heart function were significantlyimproved in the group of Fu Fang Huang Jing Oral Liquid of moderate andlarge dosage (HJm, HJh) than group ADR(P<0.01), while there was notapparently statistic difference between the groups Fu Fang Huang Jing OralLiquid moderate dosage and large dosage(HJm, HJh) (P>0.05); and there wasnot apparently statistic difference between the group Fu Fang Huang JingOral Liquid small dosage and ADR group (HJs, ADR) (P>0.05). 2. Compared with the normal control group, left ventricularhypertrophy index (LVHI) of the groups treated with Fu Fang Huang JingOral Liquid is much more than that of Group ADR(p<0.01).LVHI was significantly decreased in two treatment groups; the group ofFu Fang Huang Jing Oral Liquid of large dosage was more effective thanFu Fang Huang Jing Oral Liquid of moderate dosage; and there was notapparently statistic difference between the group Fu Fang Huang Jing OralLiquid small dosage and ADR group (HJs, ADR) (P>0.05). 3. Compared with Group ADR, the plasma and myocardial Angâ…¡, Aldconcentration decreased in Groups treated with Fu Fang Huang Jing OralLiquid, especially in the large dosage(P<0.01). 4. Compared with Group ADR, CVF both in endocardium and epicardium wassignificantly decreased in the groups treated with Fu Fang Huang Jing OralLiquid, and the effect of Fu Fang Huang Jing Oral Liquid of large ormoderate dosage group was better than that of Fu Fang Huang Jing Oral Liquidof small dosage group(P<0.01); PVCA of treated rats with drugs were lowersignificantly than that of ADR, but among the groups treated with drugs,PVCA showed no statistic significance. 5. Compared with the normal control group,the expression of myosinlight chain 2(MLC-2) in Group ADR was significantly decreased.And theexpression of MLC-2 in each group treated with Fu Fang Huang Jing OralLiquid was higher than that in Group ADR, especially the group with largedosage. 6. By ECM,better-arranged myofilament and intercalated disc inmyocardial cells, dramatical amelioration in interstitial fibrosis,congestion of blood capillary and edema of endothelial cells were observedin the groups treated with Fu Fang Huang Jing Oral Liquid.Conclusions 1. The protective effect of Fu Fang Huang Jing Oral Liquid on myocardiummay be related to their effects on the activity of RAS, as well as theirfunction of lowering MAngâ…¡,PAngâ…¡, PAld concentration. 2. Fu Fang Huang Jing Oral Liquid can decrease the plasma index ofcollagen fiber such as Pâ… NP,Pâ…¢NP,Câ…£,laminin, and thus prevent leftventricular hypertrophy and myocardial fibrosis in rats with heart failure. 3. Fu Fang Huang Jing Oral Liquid can improve the heart function byincreasing the expression of the cardiac myosin light chain 2 in rats withheart failure.
