Objcetive To study the expression and significance of three kinds of nitric oxide sythase (NOS) in renal cell carcinoma (RCC) and evaluate their roles in the growth and prognosis of tumor.Methods Iramunohistochemically staining (SABC method) was performed to detect the expression of three kinds of NOS in 60 specimens of RCC tissues and 20 specimens of normal kidney tissues for contrast. The quantitative analysis was determined by image analysis technique. Then we studied the relationship between the expression of NOS value and the tumor clinical and pathological character.Results 1. No expression of iNOS was found in the tissue specimens of two groups. 2. The expression of nNOS was mainly seen in the renal tubule cytoplasma of normal kidney tissues, and in tumor cell cytoplasma of RCC. The integral optical density (IOD) of eNOS in normal group and RCC group was 2. 22 ± 0. 22 and 2. 14 ± 0. 19 respective , P> 0. 05. The difference was not statistically significant. 3. The expression of eNOS was mainly seen in the renal tubule cytoplasma of normal kidney tissues, and in tumor cell cytoplasma of RCC .The IOD of eNOS in normal group and RCC group was 2.66 ± 0.22 and 2. 32±0. 20 respective, P <0. 01. The eNOS value in tumor tissues was much lower than that in normal kidney tissues and was negatively related to the tumor stage and grade, r=-0.823, P < 0.01 .Conclusions 1. No expression of iNOS was found in the tissue specimens of two groups. It probably was not related to the tumor genesis. 2. The difference of nNOS value in two groups was not statistically significant which suggested that nNOS might not participate in pathogenic of RCC. 3.The eNOS value in tumor tissues was much lower than that in normal kidney tissues and was negatively related to the tumor stage and grade. Which suggest that eNOS maybe play a role in the tumor genesis and development.
