Objective:As a new waveform of stimulation, chaotic electrical stimulation was used in subthalamic nucleus (STN)in epilepsy rats for a week, to evaluate the effect of STN chaotic stimulation on epilepsy and expore its potential mechanism, so provide evidence for treating pharmacological intractable epilepsy patients in such a way in stereotatic functional neurosurgery. Methods:Rats were divided randomly into three groups:stimulation group , sham stimulation group and control group. Rats in stimulation group were inserted deep electrods in STN and subdural electrod in frontal district and occipital bone' s spinous process, then delieved STN chaotic stimulation for a week, and then were provided Bemegrid (25mg/kg, intraperitonrally) to induced seizure. Ictal behaviors and synchronous electroencephalograms recordings were assessed. Sham and control groups didn' t receive any stimulation and the other treatment is the same as stimulation group. Additionly, Immunochemical (SABC) was used to investigate the alteration of GABA immuny reactive(IR) cells in neocortex in parietal lobeand hippocampus. Results:Compared with rats in control stimulation ,STN chaotic electrical stimulation significantly prolonged latency to level I (234.76 ± 54.24, 84.90 ±19.74) , level V (24. 64 ±6. 14, 8.33±6.51) and epileptiform discharge,shortened the duration of seizure after kindling(16. 23 ± 5. 49, 21.63 ±4.79), and increased the number of GABA IR positive cells of cerebrocortexand hippocampus in convulsive rats induced by Bemegrid . Sham group got thesame effect as stimulation group,but was weaker than stimulation group.Conclusion:Bilateral STN chaotic stimulation was protective against seizure induced by Bemegrid .playing a role in decreasing seizure , delaying the epileptform discharge in EEG .The effect may attribute to the effect similar to injury and enhancing the inhibitiory action of GABAergic systems in brain.
