Because the untoward reaction of chemical synthetic medicine is serious in recent years, and the Government have called on "the Chinese Traditional Medicine(TCM) Modernization",the studies of the TCM and its preparation's quality control are becoming more and more. Schisandra chinensis is one of the wide used herb of Chinese Medicine, and is a crude herb of Shengmai Injection. In order to control the quality of Schisandra chinensis and its preparation,we used protocatechuic acid as an indicative compound and developed a HPLC method to determine its content in Schisandra chinensis. Then we measured the content of 5-hydroxymethyl furfural(5-HMF) and protocatechuic acid in shengmai injection by HPLC.First we prepared protocatechuic acid reference substance for assay,and developed its quality standard. HPLC and IR were used to identify its structure,the related substance was tested by HPLC, finally we used a volumetric method to measure its content. All the results showed that the finely cleaned protocatechuic acid is fit to use as .reference substance for assay.We developed a HPLC method to determine the content of protocatechuic acid in Schisandra chinensis. Schisandra chinensis was crushed and extracted in 50%ethanol-acetic acid(100:l) by ultrasonic abstraction. We used ODS as stationary phase, mobile phase was acetonitrile-5mmol/L ammonium acetate-acetic acid(6:94:l),and the detective wavelength was set at 258nm.The method is rapid> accurate, and suitable for determination of protocatechuic acid in Schisandra chinensis.We identified a compound which was separated from shengmai injection by mass spectrum (MS)> ultraviolet spectrum (UV) and HPLC, and confirmed it was 5-HMF.Based on the previous works we develop a HPLC method to determine the content of protocatechuic acid and 5-HMF in shengmai injection. Red ginseng and ophiopogon didn't affect the accurate measurement.Protocatechuic acid and 5-HMF were good linearity when the mass is within 0.025~0.4ug and 0.05-0.8ug respectively, the correlation coefficient was both 0.999 9. and the recoveries were 99.5% and 101.3% respectively. The method is accurate, simple and durable.We investigated the source of 5-HMF in shengmai injection, and found that red ginseng and ophiopogon do not contain 5-HMF, Schisandra chinensis is also not contain 5-HMF unless it is heated in water. With the heated time increasing,the content of 5-HMF increased. We draw a conclusion that carbohydrate and glycoside in Schisandra chinensis is decomposed to monosaccharide, and then forms 5-HMF under acid condition.Usually Schisandra chinensis is used in preparation by water decoction,in the theory of Chinese Medisine, Schisandra chinensis is considered to be astringency since it is acidic. So it is. important to control the content of protocatechuic acid in Schisandra chinensis and its preparation.There is no report about the assay of organic acid in Schisandra chinensis and shengmai injection until now.
